This paper choiced the mature embryo of Xanthoceras sorbifolia as the research object, studies the generation of early mature embryo culture, cultivation of adventitious bud transgenerational and take root in the process of each influence factor, the various problems existing in the training links, and proposed the corresponding solution. The experimental results showed that:1. The effect of seeds storage temperature、time and hormone combination concentration for Mature embryo original culture.The storage of different temperature for half a year’s champions league fruit mature embryo to carry on the original culture. On the MS culture medium.-18℃storage the mature embryo of adventitious bud induction rate is the highest, at room temperature under the condition of storage rate of mature embryo induction times and the difference between the former and smaller, so considering the factors of economic and practical, choose the storage temperature of room temperature in seed storage is best. On the basis of new seeds of different storage time at room temperature at the beginning of the mature embryo of generation of cultivation experiment, only additional hormone combinations for6-BA+NAA medium can achieve a higher rate of adventitious bud induction, with the increase of storage time, additional cytokinin6-BA concentration of1.0mg.L-1medium adventitious bud induction rate is higher, and the concentration of auxin NAA as the change of storage time with no regularity.2. The influence of hormone combination and concentration of adventitious bud successive transfer culture. In the experiments of Xanthoceras sorbifolia adventitious bud transgenerational cultivation, different transgenerational number of adventitious bud adaptability to hormone concentration is slightly different,but on the MS culture medium, additional hormone combinations for6-BA+IAA+GA medium the adventitious bud proliferation effects are better than6-BA+NAA+medium of GA, and additional hormone combinations for6-BA+IAA+GA medium, no matter how long storage time, additional cytokinin6-BA concentration of0.1mg.L-1, gibberellin concentration of1.0mg.L-1on the number of adventitious bud multiplication culture medium are relatively high, the concentration of auxin IAA increased with the increase of number of transgenerational showed increasing trend. Adventitious bud late successive transfer culture,in the medium additional hormone combination 6-BA0.1mg.L-1+IAA0.5mg.L-1+GA2.0mg.L-1can improve the adventitious bud leaves tufted phenomenon.3. The effect of hormone combination and concentration of transgenerational adventitious bud rooting.In the experiment of Xanthoceras sorbifolia adventitious bud rooting, additional hormone combinations for IBA2.0mg.L-1+ABT2.0mg.L-1on the culture medium of adventitious bud rooting rate is best, can amount to50%, the average root number is3.78, root length of53.14mm; The different number of transgenerational adventitious bud of Xanthoceras sorbifolia to take root, the mix of6-BA, IAA, GA culture medium is better on the proliferation of adventitious bud its roots in6-BA, NAA. GA combination;With the increase of number of transgenerational,take root on the adventitious bud, medium additional auxin concentration should also will increase; On rooting seedlings after successive transfer culture and in the medium additional hormone6-BA0.2mg.L-1.IAA0.5mg.L-1can effectively solve the problem of above ground dry after seedling roots, and in additional hormone combinations for6-BA0.2mg.L-1+IBA0.5mg.L-1medium on successive transfer culture can also the promote.
