This paper applying the mature embryo of Xanthoceras sorbifolia as researching materials,and studying the rooting culture and somatic embryogenesis system of tissue culture system of the effects of applying different growth regulating substance on the culturing of X.sorbifolia tissue,and applying the tissue culture seedling as the further researching materials,doing a study on transplanting condition.The results are as follows:(1)By doing research on determination characteristic indices of storage time of 5 years seed,it indicated that,they doesn’t present an obvious amplitude of change on the scal index,and the rank of the thousand-grain weight index(single-grain weight index)of 5years is as follows:2012>2011>2014>2013>2015,and the thousand-grain weight index in2012 is 3.93%more than the average value(727.14g)of these 5 years.The rank of shelling percentage index is:2012>2011>2015>2013>2014,in the year of 2012,obtained the most highest shelling percentage index,and it reaches 48.28%,the most lowest shelling percentage index in the year of 2014 and it is 44.32%.(2)The materials in this study are from seeds of X.sorbifolia with different storage time,and studied the effect on callus induction from big and small cotyledon in mature embryo,the results are as those as that follow:With the seeds stored in 2011,2012,2013and 2014,the induction rate of calli induced by small cotyledon was greater than that induced by big cotyledon;the induction rate of calli induced by small cotyledon was smaller than that induced by big cotyledon in 2015.With the extension of storage time,as a regular,the induction rates of mature embryo calli decrease firstly and then rise,and the degree of rise is lower than the degree of decline,since 2012.It has been selected for five years,the induction rate of the mature embryo leaves was the highest in the explants,up to96.30%(The optimal medium is MS+6-BA 1.0 mg·L-1+2,4-D l.0 mg·L-1+NAA 0.5mg·L-1+sucrose 30 mg·L-1+agar 6 g·L-1,Culture condition:dark culturing),the calli present white,transparent,loose.Obtained the appropriate hormone concentration ratio of in the primary culturing big and small seed cotyledon of callus induction of different storage years from 2011 to 2015.(3)When doing tissue culturing research on explant by using mature embryo as material,the medium of first culturing to induction of adventitious buds is MS Medium+6-BA2.20mg·L-1+NAA0.08mg·L-1+sucrose 30g·L-1+agar 6g·L-1,pH=5.98,and the differentiation rate is 97.50%;The suitable medium for rooting culture is WPM Medium+IBA 1.0 mg·L-1+glucose 30 g·L-1+agar 5.5 g·L-1,pH=5.98,and the rooting rate can reach 86.00%,the average rooting length is 58.05 mm,the best replacement period of medium in rooting culture is 10d as a period,culture condition is seted as:3 days of no light+1 lamp of three days+2 lamps(Light intensity equal to 1644 Lux)culturing,it is effective for reducing the amount of yellow leaves produced by explants.(4)The suitable transplanting conditions for tissue culture seedlings of X.sorbifolia are:bottle opening acclimatization for 9 d,the matrix is single vermiculite,the temperature is 15-20℃,the light intensity is 2466 Lux,the survival rate of its transplanting can reach to94.46%,the growth of the seedlings was 40.5 mm,the effect of transplanting is ideal.
