| miRNA is a non-coding RNA with a length of about 18-25 nucleotides that is encoded by the body’s own genes.It plays an important role in regulating the growth and development of the body,cell proliferation and differentiation,signal transduction and immune response.In this study,a total of 9samples of skin and hair follicle tissues during the anagen(P1,May,n=3),catagen(P2,October,n=3),and telogen(P3,January of the following year,n=3)of the Hetian sheep follicles were sequenced and analyzed for the transcriptome,and the miRNAs of the same individuals with different periods of periodic changes in the Hetian sheep were screened,and the target genes were predicted by miRanda,and were analyzed by GO and KEGG enrichment for theirs function and signal pathways.The results are as follows:(1)Through the analysis of the transcriptome of the three periods of periodic changes in the Hetian sheep,143 known miRNA were identified and 759 new miRNA were predicted.173 differential expression of miRNA were found in the P2 vs P1 comparison group,of which 7 miRNA expression levels increased,The expression of 166 decreased,P3 vs P1 comparison group found 243 differential expression of miRNA,of which 28 were up-regulated and 215 were down-regulated;P3 vs P2 comparison group 71 differential expression of miRNA were found,of which 23 were up-regulated and 48 were down-regulated.The oarmiR-370-3p,oar-miR-541-3p,oar-miR-134-3p and other miRNA have significant differences in the three periods among them.This suggests that miRNA is involved in periodic changes in the hair follicle in Hetian sheep.(2)Predict target genes for the selected differential expression of miRNA,predict 1883 target genes from 173 differential expression of miRNA in the P2 vs P1 comparison group;predict 1881 target genes for 243 differential expression of miRNA in the P3 vs P1 comparison group;In the P3 vs P2 comparison group,71 differential expression of miRNA predicted 140 target genes.The target genes of KRT25 and KRT15 may be related to the development of hair follicle in Hetian sheep.(3)The predicted target genes in the P2 vs P1 comparison group was significantly enriched in 101 GO entries;the predicted target genes in the P3 vs P1 comparison group was significantly enriched in 100 GO entries;the predicted target genes in the P3 vs P2 comparison group Significantly enriched in 72 GO entries,and found that the target genes in the P2 vs P1,P3 vs P1,P3 vs P2 comparison group were annotated for hormone levels related to hair follicle development,cell catabolism,cycling binding,bone cell development,etc.In the GO entry,the predicted target genes in the P2 vs P1 comparison group was significantly enriched in 11 signal pathways;the predicted target genes in the P3 vs P1 comparison group was significantly enriched in 6 signal pathways;the P3 vs P2 comparison group was predicted to be the target genes were significantly enriched in 13 signal pathways,and it was found that they were significantly enriched in AMPK,Hedgehog,and TNF signal pathways related to hair follicle development.This study provides new information for miRNA expression profile in hair follicles of Hetian sheep by sequencing and analyzing the transcriptome of skin hair follicles during three periods of periodicity change,which is important for improving the quality of cashmere,these results provide a preliminary basis for further research on the targeting relationship of these mirnas and their target genes,and provide a scientific basis for marker-assisted selection of Hetian sheep. |
