| Hetian sheep is a fine regional breed in Xinjiang,which is one of the major economic source of livestock farmers in Hetian area.Hetian sheep also is a good breed,which can adapt low nutrition environment.The wool is famous for weaving the carpets of oriental characteristics in the world.Additionally,the domestic and foreign scholars through the research of the growth regulation of wool follicle and the regulation mechanism of the character for cashmere goat,Hu sheep,and merino sheep,they found that keratin genes become one of the important candidate gene in affecting the wool follicle development and may be has a great influence in sheep production traits.This test takes the age of 4 months Plain-type and Mountain-type Hetian sheep as research object,and taken the skin sample of abdominal lateral part in 2,4,6 and 8 months.First,the structure of the wool follicle was observed and the density of hair follicle was analyzed by making normal animal tissue sections.Second,the test used TRizol method and PCR amplification techniques of molecular biology for qualitative of the KAPs and KIFs genes(KAP1.1,KAP2.12,KAP3.2,KAP4.2,KAP5,KAP6.1,KAP7,KAP8,KIF1.2 and KIF2.9).Third,this test used the SYBR Green fluorescent dye method and real-time fluorescence quantitative technology to analyze the difference for 10 keratin genes between two kind of Hetian sheep in relative quantitative.Results:(1)Wool follicles of morphological was observed that the primary hair follicle had clearly structure and gradation,and the follicles group had obvious limit.The analysis of wool follicle density shows that the density of secondary wool follicle had little difference between Plain-type and Mountain-type Hetian sheep in different months,and the whole was increasing.However,The secondary hair follicle density of Mountain-type Hetian sheepincreased obviously from June to August.Generally speaking,secondary hair follicle density was the key stage of proliferation from April to August.(2)The positive clones of KAPs and KIFs gene of Plain-type and Mountain-type Hetian sheep was identified that we found the homologous of sequence was more than 95 %,Meanwhile,The sequence of base mutation had little effect on the composition and composition of amino acids.(3)Through quantitative analysis of 10 keratin gene shows that: There was no significant difference in KAP1.1 gene expression(P > 0.05),significant difference in KAP2.12 gene expression(P < 0.05),significant difference in KAP3.2 gene expression(P > 0.05),significant difference in KAP4.2 gene expression(P < 0.01),no significant difference in KAP5.5 gene expression(P > 0.05),and no significant difference in KAP6.1 gene expression(P > 0.05),no significant difference in KAP7 gene expression(P > 0.05),significant difference in KAP8 gene expression(P < 0.05),insignificant difference in KIF1.2 gene expression(P > 0.05),and insignificant difference in KIF2.9 gene expression(P > 0.05).Conclusion:(1)Wool follicle morphological was observed that its structure was similar to the most of animal,and the hair follicle group is mainly composed of one and two element follicle group.The density of Mountain-type Hetian sheep was slightly better than the Plaintype Hetian sheep;(2)The qualitative analysis of keratin gene confired that the 10 keratin genes exists in the sheep skin;(3)In this test,the analysis of real-time fluorescence quantitative of the two Hetian sheep showed that the differences of KAP2.12,KAP3.2,KAP4.2 and KAP8 were significant in relative expression.However,the difference in KAP4.2 gene expression is extremely significant,and the relative expression of the remaining KAP and KIF genes was not significant. |
