The aim of this study was to investigate the effects of Xiao-Jian-Zhong prescriptions and Jingsananli-sepsis on blood parameters and NF-κB signaling pathway in thymus of nursery pigs.Thirty Duroc Landrace Yorkshire nursery pigs with weight of about 17.37 ±1.32 kg and no significant individual difference were randomly divided into three groups such as Control group(Con group),Xiao-Jian-Zhong prescriptions group(TCM1 group),Jingsananli-sepsis group(TCM2 group),10 in each group(n = 10).In the control group,the piglets were fed the basic diet(without antibiotics),in the Xiao-Jian-Zhong prescriptions group,the piglets were fed the basic diet(without antibiotics)with Xiao-Jian-Zhong prescriptions,and in the Jingsananli-sepsis group,the basic diet(without antibiotics)with Jingsananli-sepsis.The experiment period is 60 d.All nursery pigs were injected with classical swine fever virus(CSFV)and porcine reproductive and respiratory syndrome virus(PRRSV)vaccines on day7 and 20 of the experiment.At 0d,30 d and 60 d,pre-vena cava blood samples were collected from all nursery pigs to determine complement(C3,C4),immunoglobulin(Ig A,Ig G,Ig M)and antibody levels(CSFV,PRRSV)in the serum.The genes and proteins associated with NF-κB signaling pathway were extracted from the thymus of nursery pigs on day 60 of the experiment.The experiment result is given as:1.Compared with the Con group,the contents of C3 and C4 in TCM1 group and TCM2 group increased significantly on the 30 th day(P<0.05 or P<0.01),while the differences of C4 in TCM1 group on the 60 th day of the experiment increased significantly(P<0.01).2.Compared with the Con group,the Ig A and Ig M contents in the TCM2 group were significantly increased on the 30 th day(P<0.01).The Ig G content of TCM2 group increased significantly on the 60 th day of the experiment and that Ig G of TCM1 group increased on the 30 th day of the test(P<0.05).3.Compared with the Con group,the CSFV and PRRSV antibody content in the TCM1 group and TCM2 group were significantly increased(P<0.01).4.Compared with the Con group,the expression levels of NF-κB signaling pathway related genes(TLR4,My D88,NF-κB,IL8 and TNF-α)in the thymus of TCM1 group and TCM2 group were significantly decreased(P<0.05 or P<0.01),but the expression levels of IL6 were not significantly different but showed a decreasing trend.5.Compared with the Con group,the expression levels of NF-κB signaling pathway related proteins(P65,p-p65,IκB-α and p-IκB-α)in the thymus of TCM1 group and TCM2 group were significantly decreased(P<0.05 or P<0.01).In conclusion,the addition of the two additives in Xiao-Jian-Zhong prescriptions and Jingsananli-sepsis may increase the level of complement,immunoglobulin and vaccine antibodies in the serum of nursery piglets,and reduce the level of thymus inflammation,so as to improve the immunity of nursery piglets.
