In order to study the effects of different hormone treatment on strawberry stem tip,anther and aseptic seedling leaf tissue culture,"Tochigii" strawberry varieties were used as test materials to establish the in vitro culture system.Through selecting and confirming the optimum hormone proportion,we studied the effects of 5 different light qualities on"Tochigii" strawberry stem tip,aseptic seedling leaf and anther culture.In addition,with"Tochigii" rootless plantlets as materials,the effects of 5 kinds of light treatments on growth of strawberry plantlets were studyed.The results are as follows:1、"Tochigii" rapid micropropagation system establishment:(1)MS+6-BA1.0mg/L+NAA0.2mg/L suitable initial culture in stem tip tissue;MS+6-BA1.0mg/L+NAA0.1mg/L for subculture,while increasing the concentration of NAA was 0.2mg/L then gradually decreasing the concentration of 6-BA from 0.5mg/L,0.25mg/L to0mg/L,suitable for the growth of strawberry plantlets.in vitro culture medium for rooting was MS.(2)The combination use of Tween-80 and the 75% alcohol was beneficial to reduce the contamination rate of anther inoculation."Tochigii" initial medium MS+6-BA1.5mg/L+NAA0.2mg/L+2,4-D0.1mg/L is most suitable for the anther callus induction;MS+6-BA2.0mg/L+NAA0.1mg/L+2,4-D0.05mg/L is the most suitable differentiation medium for anther callus,culture medium for rooting was MS.MS+TDZ2.0mg/L+2,4-D0.1mg/L culture aseptic seedling leaf tissue suitable for callus induction.In subculture stage,MS+TDZ2.0mg/L+2,4-D0.05mg/L;MS+TDZ1.0mg/L+2,4-D0.2mg/L was proliferation medium;2,4-D concentration was 0.2mg/L,the concentration of TDZ was gradually decreased from 2.0mg/L to 1.0mg/L,0.5mg/L,0mg/L can significantly improve the seedling growth,culture medium for rooting was MS.2、The effects of light quality on "Tochigii" strawberry tissue culture:(1)The strawberry shoot tip differentiation rate was the highest under white light treatment.proliferation coefficient was the highest under Red /Blue/Far Red=2:1:1treatment.Under the treatment of red light,shoot differentiation was the earliest and the growthof buds differentiation was fastest.On rooting stage,under the treatment of Red/Blue/Far Red=2:1:1,the rooting rate and root vigor stand at the highest value.(2)During the induction of anther callus,under Red/Blue/Far Red=2:1:1 treatment,callus induction rate was highest and grew well;Under red light treatment,the callus germination was the earliest;Under white light treatment,the callus differentiation rate was the highest.(3)In the process of induction of aseptic seedling leaves,the red light was beneficial to the induction of callus,and the proliferation coefficient was the highest under the white light treatment.The rooting rate under Red/Blue/Far Red=2:1:1 was the highest,and the plant growth vigor was strong.(4)By testing the content of chlorophyll and enzyme activity of the aseptic seedling leaf,anther callus,the results showed that the content of chlorophyll in anther callus was highest under the white light,the differentiation rate was significantly greater than the other treatment;blue light treatment significantly improved the content of soluble protein in callus;the water content of callus under was the highest under red light treatment;Under Red/Blue/Far Red=2:1:1 treatment,the SOD,POD,CAT activity reached the maximum value and the callus grew well.3、The effects of light quality on the growth of strawberry plantlets:Under blue light,the growth of plants was most vigorous,the leaf area was big,the soluble protein content of leaves was the highest,but the root is poor;the content of soluble sugar and CAT activity under white light treatment were the highest;Under Red/Blue/Far Red1:2:1 treatment,the activities of SOD and POD enzymes were the highest the plants grew fast and training time is shortened. |
