In this experiment,“Xiang Ye”strawberry tissue culture seedling was used as test.material the optimum plant growth inhibitor suitable for in vitro preservation of germplasm resources of strawberry in vitro culture seedlings,and the optimum concentration,as well as the effect of light intensity on the preservation of strawberry seedlings were studied.The test results are as follows:1.Selection of optimal medium osmotic pressure for in vitro preservation of strawberry tissue culture seedlings:Comparison of growth and preservation of“Xiang Ye”strawberry tissue culture seedlings by adding different regulatory substances to the medium to change the medium osmotic pressure Sucrose concentration of 60 g/L somaclone survival rate was 83.7%,the medium to40 g/L’s survival rate is 90.7%when the concentration of mannitol,inositol best of 0.2 g/L under the strawberry seedlings survival rate is 70.2%,it is strawberry somaclone short-term save medium for MS+NAA0.2mg/L+6-BA0.5mg/L+Mannitol 40g/L+Sucrose 30g/L+Agar7g/L,about p H5.8,day light intensity is 60μmol·(m~2·s)-1,control temperature is 25℃.2.Selection of optimal plant growth inhibitors for in vitro preservation of strawberry tissue culture seedlings:Comparison of growth and preservation of strawberry tissue culture seedlings by adding different types and different concentrations of plant growth inhibitors in the medium:Within the 150 d and other conditions of the same cases,abscisic acid(ABA)concentration in culture medium for 6 mg/L maximize survival rate 87.3%,add 35 mg/L of cycocel(CCC)survival rate of 86.3%,when the medium multi-effect azole(PP333)concentration of 1.2 mg/L of survival rate is 80.6%,the medium add 20 mg/L of butyl hydrazine(B9)survival rate is 80.4%,1.7 mg/L green fresh element(MH)effect is poorer.It is“Xiang Ye”strawberry somaclone save medium MS+NAA0.2mg/L+6-BA0.5mg/L+ABA6mg/L+Sucrose30g/L+Agar7g/L,Other environment unchanged.3.Selection of optimum light for in vitro preservation of strawberry tissue culture seedlings:By comparing the growth and preservation of strawberry tissue culture seedlings under different light conditions,it was found that the light intensity of 9mol·(m~2·s)-1was the optimum light intensity for in vitro preservation of“Xiang Ye”strawberry.However,the survival rate of“Xiang Ye”strawberry seedlings could not be guaranteed by adjusting the light intensity alone,and the survival rate of tissue culture seedlings was 65.3%.
