Classical swine fever(CSF)is a kind of contact,high fever and hemorrhagic infectious disease caused by classical swine fever virus,the World Health Organization(OIE)included classical swine fever in a class A of infectious disease,China is also included it in a class of animal disease.As one of the most serious infectious diseases of swine industry in china,it has seriously restricted the development of pig industry.Purpose:Immune monitoring the CSFV antibody of pig in a large scale pig farms in West Fujian,detecting the status of virus carried by classical swine fever virus of Sow umbilical cord blood,analysis the genetic variation of E2 gene of classical swine fever virus,In order to master the immune effect of classical swine fever and the genetic changes situation of classical swine fever virus,to provide a theoretical basis for the prevention and control of hog cholera.Method:This research method mainly used the detection means of ELISA and PCR real-time,RT-PCR,detected the swine serum and CSF pathogen of this field scale.Results show:1)The monitoring results of the swine fever antibody in the field between 2014 and 2015,the different stages of swine swine fever antibody qualification rate between 77.3%-100%in 2014,the positive rate in delivery room was the highest 100%piglets swine fever antibody,the second is the reserve of pigs,pigs,pigs and piglets,pass rates were 93.4%,90%,84.1%and 73.3%.The different stages of swine swine fever antibody qualification rate between 34.3%-96.8%in 2015,the positive rate of swine fever antibody reserve up.to 96.8%,the second is the pig,delivery room pigs,piglets,Pass rates were 90%,83.9%,79%and 34.3%,According to the different stages of pregnancy to make sow classification,divided into pregnant 30-45d,pregnant 60d,pregnant 90-100d,four types of lactation sows,the average blocking value of swine fever antibody were 71.5%,76.7%,72.7%,72.3%,discrete degree was 24.2%,21.1%,20.9%,24.8%.According to the different parities to classify tested sows serum samples were classified into 1 parity,2parity,3 parity,4parity,5 parity,6 parity and above parity,the average blocking value of swine fever antibody were 73.9%,71%,74.4%,75.1%,76.3%,67.8%,Eligibility rates were 82.1%,83.3%,88%,100%,94.4%,87.5%.2)Comparison of 2 different immunization program,the results show that the difference of the average classical swine fever antibody blocking value level in the 2 kinds of immunization programs in 2W,4W,8W,10W,12W is not obvious,in section 6w,the swine fever antibody blocking value level in the 2 immunization program has increased by nearly 30%than 1 groups of immune procedures.3)Detection of umbilical cord blood of swine fever disease,the detection score was 117 and the positive rate was 9,the positive rate was 7.69%.Analysis of 5 positive sample of classical swine fever sequencing and drawing evolution tree by sequence alignment,showed that CSFV E2 nucleic acid sequence for genotype 1.1 and HCLV strain had the highest homology reached more than 96.3%.Conclusion:the conclusion is that the field of CSF had a good immune in 2014 and 2015,the differences in immune effect of commercial pig is big,the conservation of swine fever antibody level is the worst.The differences of sow CSF antibody level in different stages of pregnancy is not obvious.Different Parities have influence on the mother of swine fever antibody level,after fifth birth,the CSF antibody and qualified rate had decreased.Compare 2 different immune procedures,this field application of immune procedure 2 has more clinical value.Detection of classical swine fever and the umbilical cord blood of sows CSFV E2 gene sequencing,proved that the tested strains were genotype 1.1 and the homology of HCLV strains reached more than 96.3%.
