Fruit of Rosa davurica Pall.has a wide range of pharmacological activities owing to rich in a variety of chemical components which has great potential in the field of new drug development and new functional food.Due to the limitation of modern separation technology,there is no significant breakthrough in the study of structure-activity relationship between monomer components and pharmacological activities in the fruit of Rosa davurica Pall..The chemical constituents and bio-activities in the fruit of Rosa davurica Pall.were studied which provides an important theoretical and experimental basis for the development of fruit of Rosa davurica pall.related products in the future.The main research results of this paper are as below:1.The preparation of total flavonoids-Ca2+complex of fruit of Rosa davurica pall.has opened up the way for its further comprehensive utilizationDynamic adsorption-desorption method of macroporous adsorption resin was used to purify the total flavonoids of fruit of Rosa davurica pall.extract with the content of total flavonoids more than 60%.Using the purification as raw material,the best preparation process of total flavonoids of fruit of Rosa davurica pall.-Ca2+complex were optimized:Using anhydrous methanol as solvent,the mass concentration of the extract was 0.72 mg/ml,the complexing time was 4 h,the water bath temperature of the constant temperature oscillator was 30℃,the mass concentration of Ca Cl2 was 6 mg/ml,and the p H value of the system was 10.5,which offer the complexation conversion 98.43%and the final yield 65.48%.So far,there is no relevant report about this study which provides a research basis for finding new,efficient and low toxic flavonoids metal complex drugs.2.The relationship between different chemical components in the fruit of Rosa davurica Pall.and their bio-activities was studied qualitatively and quantitativelyPetroleum ether,ethyl acetate and water saturated n-butanol were used to extract the total flavonoids of fruit of Rosa davurica Pall.in turn,and the extraction phases with different polarity were obtained,UV-vis method and HPLC method were applied to study the content determination and bio-activities analysis of the active components in different polar phases.Using crude extract of fruit of Rosa davurica Pall.as reference group,its biological activity was evaluated by scavenging·OH,DPPH·,nitrite,anti-liposome peroxidation and FRAP method.The results show that most polarity substances were found in the fruit of Rosa davurica pall.,most of which are enrich in ethyl acetate extraction phase and n-butanol extraction phase.secondly,and all phases showed strong biological activity.The contents of flavonoids,saponins,polyphenols and tannins were correlated with the scavenging ability of DPPH·,·OH,nitrite,the anti-lipid peroxidation activity and FRAP in varying degrees.At present,there is no relevant report on fruit of Rosa davurica pall..This study provides material information for further study on the biological activity in the fruit of Rosa davurica pall..3.The chemical constituents of n-butanol extraction phase of fruit of Rosa davurica pall were separated by various methodsTwenty-monomer compounds were isolated from the n-butanol extraction phase of fruit of Rosa davurica Pall.by silica gel column chromatography,reversed-phase C18column chromatography,dextran gel column chromatography,semi-preparative HPLC,recrystallization and other advanced separation technology.The structures of twelve compounds were identified,among which six compounds were identified by nuclear magnetic resonance(1HNMR,13CNMR).They are 5-ethoxy-2-methoxy methyl-6-(1-methyl-butoxy)-tetrahydropyran-3,4-diol(C1),6-(hept-5-ene-3-yl)oxy)-2-(methoxy methyl)-tetrahydropyran-3,4,5-triol(C2),3-methyl-hexane(C3),acetic acid(C4),Ethylene glycol acetate monoester(C5),Rhamnose(C6),including 2new compounds(C1,C2).Six known compounds were identified as rutin(C7),quercetin(C8),oleanolic acid(C9),ursolic acid(C10),proanthocyanidin B2(C11)and gallic acid(C12)by HPLC.4.The effects of simulated gastrointestinal digestive juice on the active components and antioxidant activity in the fruit of Rosa davurica pall were studiedBased on the study of chemical components,the absorption of functional components in the fruit of Rosa davurica Pall.after body ingestion was investigated from the perspective of dynamics.Eight active components and antioxidant activities of crude extract from fruit of Rosa davurica pall.were studied by simulated digestion in vitro.The results indicated that there were significant differences in the contents of active components before and after digestion(P<0.05).The scavenging capacity of·OH,ABTS+and superoxide anion radical of the products after digestion showed a decreasing trend after gastrointestinal digestion.The functional components and bio-activities showed a decreasing trend after gastrointestinal digestion,among which the oral and gastric digestive juices had the strongest antioxidant capacity,while intestinal dialysate had the weakest antioxidant capacity,which provided a scientific basis for studying the metabolic kinetics of active components in the fruit of Rosa davurica pall..
