The fruits of Rosa davurica pall.(FRDP)is the mature fruit of rose thorn which is regarded as natural antioxidants owing to its rich chemical components.As a Chinese herbal medicine it is allowed to be used in health food.Using aqueous extracts from FRDP as main research objectives,first of all,the contents of main functional components were determined and the activity in vitro and stability of antioxidant activity were investigated;the second,the chemical components of FRDP were isolated and identified in order to explore the relationship between its activity and chemical substances,which may provide basic data for further utilization and development of FRDP;the last,the health food-Ciqishen capsule was preliminarily studied on the basis of the previous stage.The content of functional components in aqueous extracts from FRDP was determined.The total flavonoids content was 75.33mg/g by Na NO2-Al(NO33-Na OH chromogenic method;the total polyphenol content was 173.00 mg/g by folinol chromogenic method;the proanthocyanidins content was 139.02 mg/g by ammonium ferric sulfate chromogenic method;the content of anthocyanin was 6.65 mg/g by sodium sulfite bleaching method.The results showed that there were a large number of functional components in aqueous extracts from FRDP which account for about 40%of the total.The activity in vitro of the aqueous extracts from FRDP was studied.The results indicated that it had good whitening effect in vitro,had obvious inhibition effect on tumor cells and adenase,had hypoglycemic and lipid effects,had weak inhibition effect on uric acid.In addition,it also showed significant antioxidant effect.The stability test results of antioxidant capacity showed that the introduction of sodium ions,potassium ions,calcium ions,zinc ions and copper ions had little effect on the antioxidant capacity.The anti-oxidation ability was not significantly affected by the conditions of sunlight,indoor light,incandescent light,ultraviolet dark light and light avoidance.In different temperature influence test,the scavenging ability of DPPH free radical decreased obviously when the temperature reached 100℃.In different p H value influence test,the scavenging ability of DPPH free radical decreased when the p H value reached 10.The chemical constituents of the aqueous extracts from FRDP were separated by column chromatography,centrifugal TLC and TLC.Twelve components were isolated and analyzed by gas chromatography-mass spectrometry and ULTRA-performance liquid chromatography-tandem quadrupole time of flight mass spectrometry.Eleven components were deduced,which provided chemical basis for further study of their efficacy factors.According to the above research results,FRDP,ginseng and astragalus membranaceus were developed for health food which could enhance immunity.The preparation of capsule and the detection method of signature ingredient of polysaccharide and saponin were studied.Through systematic study the aqueous extracts from FRDP,such as,the analysis of active components,the activity test in vitro,the separation of components and the development of health food,which provides support for the further comprehensive utilization of FRDP.
