| Being a natural resource with great economic value,Fruits of Rosa davuvrica Pall.was studied to provide scientific evidence for its development and utilization.The extraction and purification methods of total saponins in fruits of Rosa davuvrica Pall.and its activity in vitro were studied in detail.First,the extraction methods of total saponins in fruits of Rosa davuvrica Pall.were optimized.Many extraction methods,such as reflux,microwave,enzyme,microwave coenzyme and microwave synergistic surfactant extraction were applied to extract the total saponins from the fruits.Using oleanolic acid as reference substance,the concentration of total saponins in extracts was determined by ultraviolet-visible spectrophotometry method.Applying extraction efficiency of total saponins for index of evaluation,the optimal extraction condition was decided according to the comparison of different methods.Second,the purification methods of total saponins in fruits of Rosa davuvrica Pall.were optimized.Eight kinds of macroporous resins,such as LSA-21,LSA-10,LSA-33,ADS-17,AB-8,D-101,ADS-F8 and LX-36 were used to purify the total saponins.The adsorption capacity and the desorption capacity of total saponins were decided with ultraviolet-visible spectrophotometry method.The desorption rate of total saponins was used as a index to decide the better purification conditions.Finally,the antioxidant and anti lipid activity of total saponins from fruit of Rosa davurica Pall.were investigated in vitro,and the antiglycemic activity in vitro was evaluated by the inhibitory effect of total saponins extract on alpha glucosidase.The optimum extraction method of total saponins from fruit of Rosa davurica Pall.was microwave combined with enzyme extraction.The optimal technological conditions were obtained as followed.The ratio between material and solvent,enzyme hydrolysis time,enzyme hydrolysis temperature,volume fraction of ethanol,microwave duration,water bath temperature and microwave power were 1:36,2 h,70 ℃,55 %,8 min,60 ℃and 600 W,which might offer the extraction efficiency of total saponins to 3.15 %.LSA-21 macroporous resin has the best purification effect on total saponins.The optimum conditions were as follows.The concentration of the adsorbate,the volume of the adsorbate,the adsorbate p H value,the volume fraction of ethanol,the solution volume and the solution absorption p H value were 0.1927 mg/m L,25 m L,6.5,90 %,40 m L and 8.5 respectively.Under the optimal purification conditions,the adsorption rate and desorption rate of total saponins reached 77.51 % and65.28 %,and the concentration of total saponins in dry extract could be increased from 9.49 % to33.65 %.The scavenging action of purified total saponins from fruit of Rosa davurica Pall.to DPPH,OH,superoxide anion free radicals and ABTS radicals could reach 86.09 %,69.05 %,54.60 % and34.58 %,and the inhibition rate to lipid and alpha glucosidase could reach 70.24 % and 71.53 %respectively.Microwave combined with enzyme method has better extraction effect on the extraction of total saponins from fruit of Rosa davurica Pall.and the extraction rate can reach 3.15 %.After being,the content of total saponins from fruit of Rosa davurica Pall.in dried extract increased from 9.49 % to 33.65 %.The total saponins extracted from fruit of Rosa davurica Pall.and purified by LSA-21 macroporous resin had certain antioxidant and hypoglycemic activities in vitro which showed good development and utilization potencial. |
