The fruits of Rosa davurica pall.(FRDP)is the mature fruit of the rose thorn rose of the family Rosaceae.The dry roots and rhizomes of the Acanthopanax senticosus and ginseng are the Araliaceae plants.The three are rich in active ingredients such as saponins,polysaccharides and flavonoids,etc.They belong to Chinese medicinal materials which can be used in health foods owing to their strong anti-fatigue effects.Therefore,three of them were used as raw materials to prepare a health food with anti-fatigue function-Shuangcishen Capsule.The preliminary study on both extraction and purification process and also the formulation of quality standards were conducted.First of all,the content of flavonoids in the extract of FRDP was studied.High performance liquid chromatograph(HPLC)was used to establish a method for the simultaneous determination of the contents of four flavonoid monomers in the extract of FRDP.The contents of hyperoside,rutin,quercetin,and luteolin in the extracts were 607.98,450.63,187.32,12.68μg/g.According to the results of content determination,the compound compatibility of 4flavonoid monomers was designed,the the anti-oxidation,anti-tumor and hypoglycemic activities of the extracts of FRDP,four flavonoid monomers and their combination were investigated respectively.In the in vitro antioxidant activity test of each test article,the activity of quercetin is stronger,and the activity of the compound is weaker than that of the extract of FRDP.In the in vitro anti-tumor experiment,luteolin has better nitrous acid salt removal activity,blocking the compound compatibility in the nitrosamine synthesis experiment is stronger than the monomers themselves.And the activity experiment of inhibitingα-glycosidase in vitro,quercetin has a stronger activity.Secondly,the extraction process of Shuangcishen Capsule was screened.The process conditions of ethanol hot reflux method,column chromatography circulation combined extraction method and natural eutectic solvent cooperative microwave-ultrasonic extraction method were optimized.Under the corresponding optimal extraction conditions,the total saponin extraction rates of the three extraction methods were 28.84,29.02 and 48.28 mg/g,respectively.Then the macroporous resin static adsorption-desorption method was applied to purify the total saponins in the extract of Shuangcishen.Single factor inspection was conducted using adsorption rate and desorption rate as inspection indexes.The process was optimized using method of orthogonal experiment.Under optimal condition,the purity of total saponins in the extract of Shuangcishen Capsules was increased from 17.83%to 37.04%.Ultraviolet spectrophotometry and HPLC were used to establish the determination method of total saponins and ginsenoside Rg1,Re,Rb1 and syringin in Shuangcishen Capsules.The total saponin content in Shuangcishen Capsule was 13.32 g/100g,and the contents of four anti-fatigue functional factors ginsenoside Rg1,Re,Rb1 and syringin were 137.90,189.58,356.83,114.99 mg/100g,respectively.Finally,the activity of the contents of Shuangcishen Capsules was studied in vitro,which showed that Shuangcishen Capsules had certain antioxidant and antitumor activities in vitro.To sum up,the content and activity in vitro of flavonoids in extract of FRDP,the preparation process and quality standards of the health food Shuangcishen Capsules were systematically analyzed and studied.The research provided a strong scientific foundation and study basis for the comprehensive utilization and development of Real estate Chinese herbal medicine such as FRDP,ginseng and acanthopanax senticosus of Jilin Province.
