Lipidomics,an emerging experimental technique,is utilized to identify and quantify the endogenous lipid metabolism and investigate the signal of lipid metabolic pathway.In this way,lipidomics has played a crucial role in disease diagnosis or treatment and the biomarkers identification of disease and so on.There are two methods involved in lipidomics,including targerted lipidomics and untargerted lipidomics.And the untargerted lipidomics would investigate all of the endogenous lipid metabolites in the biological system,so that the untargerted lipidomics is more suitable for the identification of biomarkers.Fatty acids,as one of important components of lipid,are characterized into three types according to the number of double bonds:saturated fatty acid(SFA),monounsaturated fatty acid(MUFA),polyunsaturated fatty acids(PUFA).Among them,PA,the representative SFA,and OA,the representative MUFA,could regulate the biological activity of hepatocytes based on kinds of mechanism.However,there was no research that investigated the endogenous lipid metabolites influenced by PA or OA administration in hepatocytes based on lipidomics until now.Therefore,HPLC-MS-based untargeted lipidomics was used to identify and relatively quantify the endogeous lipid metabolites with PA or OA administration in this study,and investigate the tendency of lipid metabolites after PA or OA treatment.As results showed,the endogenous lipid metabolites were both significantly altered after PA and OA treatment,and the identified lipid metabolites were characterized as 22 lipid classes and they were further grouped into the 4 categories including phospholipids,sphingolipids,neutral lipids and 'fatty acyl and other lipids'.After statistical analysis,there were 12 lipid classes presented significant difference after PA treatment,including increased lysophosphatidylcholine(LPC),phosphatidylinositol(PI),sphingosine(SPH),ceramides(Cer),hexosylceramides(HexlCer),diglyceride(DG),triglyceride(TG),acyl carnitine(AcCa)and N-acylethanolamine(AEA),and decreased dihexosylceramides(Hex2Cer),sphingomyelin(SM)and cholesterol ester(ChE).At the same time,there were 10 lipid classes were significantly altered after OA treatment,among them,LPC,phosphatidylglycerol(PG),Cer,Hex1Cer,Hex2Cer,ChE and coenzyme(Co)were obviously reduced,and DG,TG and AcCa were significantly accumulated.In summary,HPLC-MS-based untargeted lipidomics was used to identify and relatively quantify the endogenous lipid metabolites with PA and OA administration,respectively,and the results of multiple analysis and statistical analysis proved that the endogenous lipid metabolites were significantly changed after PA and OA treatment.
