Object:Endometriosis(EMT)refers to the appearance of endometrial tissue(glands and stroma)outside the uterus.It occurs in women of childbearing age,and most commonly occurs in ovaries and uterus.ligament.Endometriosis is benign in morphology,but has clinical behavioral characteristics similar to malignant tumors,and has a certain malignant tendency.Comprehensive studies have shown that its malignant rate is about 0.7 to 1%.The purpose of this study is to preliminarily study the genes related to the pathogenesis of endometriosis-associated ovarian clear cell carcinoma,and provide data for finding meaningful markers and target genes for targeted therapy.Method:Search and download the original data in the GSE7305 and GSE6008 datasets from the GEO database,use R language to normalize the data and then obtain the differential genes between the diseased tissues and normal tissues in each dataset,and take the intersection of the two sets of differential genes,and Perform enrichment analysis on the intersection genes,and screen out the pathways and genes that may be meaningful for discussion and analysis..Select more stringent screening conditions for differential gene screening and enrichment analysis,and select two genes with larger expression differences and the pathways involved in these two genes to analysis.Application of immunohistochemistry to detect the expression of selected genes in paraffin specimens of endometriosis-associated ovarian clear cell carcinoma,non-endometriosis-associated ovarian clear cell carcinoma,ovarian endometriosis,and normal ovarian tissue,and conduct statistical analysis.Result:Using P value<0.05,log?2|Fold Change|>1 as conditions,the differential genes were screened out,and there were 610 overlapping genes in the two data sets.After enrichment analysis,it can be seen that these differential genes may affect p53 and other pathways,and may also affect various biological processes in ovarian cancer cells.After selecting more stringent screening conditions,143 different genes were screened out.After selecting more stringent screening conditions,and carry out enrichment analysis,select RGS2,MYLK gene and c GMP-PKG pathway for discussion and analysis.The expression of RGS2 and MYLK in endometriosis-associated clear cell carcinoma of the ovary was verified by immunohistochemistry on paraffin specimens.The results showed that RGS2 was associated with endometriosis-related clear cell carcinoma of the ovary.There are differences in expression of non-endometriosis-associated ovarian clear cell carcinoma.But there was no statistically significant difference in the expression of MYLK between the endometriosis-associated ovarian clear cell carcinoma group and the non-endometriosis-associated ovarian clear cell carcinoma group..Conclusion:1.Based on bioinformatics analysis,the expression changes of RGS2,MYLK,CEACAM1,VEGFA,EGFR and other differential genes may be involved in the malignant process of ovarian tissue.Among the differential genes whose expression is down-regulated,the difference between RGS2 and MYLK is particularly significant.2.The expression of RGS2 is down-regulated in endometriosis-associated ovarian clear cell carcinoma.3.Although MYLK showed a trend of down-regulation in the endometriosis-associated ovarian clear cell carcinoma group,there was no statistically significant difference between the non-endometriosis-associated ovarian clear cell carcinoma group.
