Objective:Recent scientific studies have confirmed that SPC24,a subunit of Ndc80 complex,plays an important role in the occurrence and development of tumors.However,the relationship between SPC24 and renal clear cell carcinoma has not been reported,and its potential role in the occurrence and progression of renal clear cell carcinoma needs to be further explored.The purpose of this study was to explore whether SPC24 can be used as a biomarker to predict the progression of renal clear cell carcinoma and its relationship with immune microenvironment and pan-cancer.Methods:The genomic and clinical data of patients with renal clear cell carcinoma were collected from The Cancer Genome Atlas database(TCGA database)and the Gene Expression Omnibus database(GEO database),and the data were processed by R software.Finally,the SPC24 gene expression information and related clinical information of all patients with renal clear cell carcinoma in TCGA and GEO database were collected.Then,we analyzed the difference of SPC24 expression between renal clear cell carcinoma and normal renal tissue.Then,according to the median of SPC24 expression,patients with renal clear cell carcinoma were divided into high expression group and low expression group,and the differences of overall survival,disease-free survival,progression-free survival and disease-specific survival between high expression group and low expression group were compared by survival analysis.We also evaluated the relationship between the expression of SPC24 and clinicopathological stage by clinical correlation analysis.Then,we performed univariate Cox regression analysis and multivariate Cox regression analysis to compare the relationship between SPC24 expression and other clinical features and the overall survival of patients with renal clear cell carcinoma,and to explore whether SPC24 can be used as an independent prognostic index to predict the prognosis of patients with renal clear cell carcinoma.After that,we verified the expression of SPC24 in renal clear cell carcinoma cell lines(OSRC,786O)and normal renal cell lines(HK2)by RT-q PCR analysis.In addition,in order to identify the signal transduction pathways related to SPC24 expression,we also carried out gene enrichment analysis(GSEA).We also used CIBERSORT algorithm to study the proportion of immune cell infiltration in renal clear cell carcinoma to further clarify the correlation between SPC24 expression level and immune microenvironment.Finally,in order to explore the potential prognostic value of SPC24 in pan-cancer,we further studied the relationship between SPC24 and 32 other cancers.Results:Compared with normal renal tissue,SPC24 was significantly overexpressed in renal clear cell carcinoma.Compared with patients with low expression of SPC24,patients with high expression of SPC24 had significantly poorer overall survival,disease-free survival,progression-free survival and disease-specific survival.After clinical correlation analysis,we found that the expression level of SPC24 was positively correlated with the histological grade,pathological stage and TNM stage of patients with renal clear cell carcinoma.Multivariate Cox regression analysis showed that the expression of SPC24 could be used as an independent risk factor for predicting the prognosis of patients with renal clear cell carcinoma.The results of PCR assay showed that the expression of SPC24 in renal clear cell carcinoma cell line(OSRC,786O)was significantly up-regulated compared with HK2 cell line in normal renal tissue.In order to predict the prognosis of patients with renal clear cell carcinoma more effectively,based on clinical indicators(age,sex,histological grade,clinical stage)and SPC24 expression,we established a line chart to predict the 1-,3-and 5-year survival rates of patients with renal clear cell carcinoma.Through GSEA analysis,we found that many pathways were significantly enriched in the high expression group of SPC24,including many tumor-related pathways and immune-related pathways.In addition,further analysis of pan-cancer data showed that the expression level of SPC24 in a variety of tumor tissues was significantly increased,and the higher the expression level,the worse the prognosis of patients.Finally,through Ciber Sort algorithm analysis,we found that in renal clear cell carcinoma and 32 kinds of pan-cancer tissues,the expression of SPC24 can significantly affect the proportion of immune cell infiltration in tumor microenvironment.Conclusions:SPC24 is a biomarker that can predict the prognosis of patients with renal clear cell carcinoma,and is closely related to immune microenvironment,so it has popularizing value in pan-cancer. |
