In this paper,ubiquitin ligase(BTRC)feedback/up/downstream activation and inhibition molecular knowledge computing network was constructed in human left hemisphere,which is closely related to cognition by integrating GRNInfer and DAVID.Based on BTRC feedback/upstream/downstream activation and inhibition common knowledge of different molecules,BTRC new systemic mechanisms are proposed and verified for regulating human left hemisphere cognition:Integrating BTRC upstream-activatd,downstream-activating Alzheimer's disease,the corresponding upstream-activates platelet activation,G Protein Signaling | Oocyte meiosis | phosphatidylinositol binding | post embryonic development | stimulatory C type lectin receptor signaling pathway,epidermal growth factor receptor signaling pathway,fibroblast growth factor receptor signaling pathway,Fc epsilon receptor signaling pathway,small molecule metabolic process,innate immune response,vascular endothelial growth factor receptor signaling pathway| microtubule binding,negative regulation of intracellular transport,positive regulation of axon extension,mitochondrion transport along microtubule,axon extension,microtubule cytoskeleton | enzyme binding,brain development,plasma membrane | apoptotic process,programmed cell death in ITPR1,SEL1L,MAPT,PRKCI2,CDR2,TULP3,ANAPC10,AL109696,DICER1 subnetworks;or downstream-activates respiratory electron transport chain,cellular metabolic process,Oxidative phosphorylation,Parkinson's disease,Huntington's disease,primary active transporter activity,inner membrane,monovalent inorganic cation transporter activity,mitochondrial inner membrane,mitochondrial membrane,hydrogen ion transporter activity,electron transport Chain in PRKCI1,HSP90AB1,R37702,NDUFA5,ATP5J GTF2I1 subnetwork.BTRC is self feedback activating Alzheimer's disease affects cognition through platelet activation | Oocyte meiosis |phosphatidylinositol binding | post embryonic development | innate immune response | microtubule binding | brain development | apoptotic process | hydrogen ion transporter activity for positive varificationBTRC mainly positive feedback inhibits DNA binding and metal ion binding.It is proposed that BTRC positive feedback inhibits DNA binding and metal ion binding,and it negatively regulates the transcription of RNA polymerase ? promoter activated by positive feedback.It is verified by inhibiting gene expression and its upstream activated or feedback inhibited mRNA processing reactants for mutually verification.Through the negative feedback activation or inhibition of enzyme binding,endocytosis,positive regulation of GTPase activity,gene expression,signal transduction and small GTPase mediated signal transduction of BTRC,it is proposed that BTRC affects cognition through speed limiting signal transduction and endocytosis for negative varificationIn conclusion,the above new hypothesis is put forward and verified.The new systematic analysis of BTRC adds theoretical and applied research on human left hemisphere cognition,and provides important theoretical basis and application value for the study of human left hemisphere cognitive mechanisms.
