Climate change is regarded as the biggest threat to ecosystem and biodiversity.However,the impact of climate warming on the gut microbiota and immunity of wild animals,especially reptiles,is largely unknown.The gut microbiota and immune ability of animals play an important role in food digestion,nutrition absorption,energy metabolism,disease prevention and behavior,which is very important for their survival and reproduction.Therefore,in this study,the control group and the heating group(+5.5?)were set up to analyze the effects of heating on the microbial structure,microbial diversity and cellular immunity of Eremias argus by high-throughput sequencing,leukocyte count and PHA cell immunoassay.The purpose of this study is to explore the impact of temperature rise on the physiological function of reptiles under the background of global climate change,and to provide scientific guidance for the protection and management of reptiles in the future.The main results are as follows:1.A total of 18 439 OTU were detected in the fecal samples of Eremias argus,9 493 in the heating group and 5 495 in the control group,with a total of 3 435 OTU in both groups.There were 32 phyla,252 families and 492 genera in the control group,and 30 phyla,225 families and 424 genera in the heating group.2.The core phylum of gut microbiota in Eremias argus is Bacteroidetes,Firmicutes and Proteobacteria,but the proportion of each micro flora was different in each group.3.Alpha diversity analysis showed that the richness and diversity index of Intestinal flora in the control group were significantly higher than those in the warming group(P<0.01).Comparing the two groups of different genders,it was found that the bacterial diversity of male lizard in the heating group was significantly lower than that in the control group(P<0.05),and the bacterial richness of female lizard in the heating group was significantly lower than that in the control group(P<0.01).Bate diversity analysis of gut microbiota showed that the isolation of intestinal microflora in the two groups was not obvious,and the similarity of Intestinal flora in the control group was lower than that in the warming group.4.Comparing the difference of the two groups of bacterial flora,it was found that the intestinal flora of the control group had 3 classes,3 orders,10 families and 13 genera,and the warming group had 1 class,1 order,6 families and 6 genera.The abundance of pathogenic bacteria such as Acinetobacter and Anaerotruncus in the intestinal flora of Lizard lizards in the warming group was higher.5.The number of leukocytes of Eremias argus ranged from 5 120 to 13 600/mm3,which was significantly different between the two groups(P<0.05).The number of leukocytes of female Eremias argus decreased more than that of male after heating treatment.6.PHA lymphocyte proliferation test showed that the response time of lymphocyte proliferation was prolonged and the proliferation degree was weakened in the warming group,but there was no significant difference between the two groups.Conclusion:high temperature can significantly reduce the richness and diversity of gut microbiota,reduce the number of leukocytes and the ability of lymphocyte proliferation and differentiation.That indicated high temperature has adverse effects on intestinal microflora and part of immune ability of Eremias argus,especially on the female.
