Mitochondrial DNA(mitochondrial DNA, mtDNA) is used as an independent of the nuclear genome of organelles, with maternal inheritance, independent restructuring and other characteristics, Nearly 20 years, one of the most important molecular marker is marker of mitochondrial genes, which are widely used in various aspects of molecular ecology, in terms of system status determination system, the relationship between animal groups and has played a positive role(Boore and Brown,1998; Carapelli et al., 2007). Currently, the measurement and analysis of mitochondrial genome sequence and to cause the molecular systematics researchers with great interest as molecular markers have been, and gradually become the main means of genetics and molecular genetic study of animal evolution.In this thesis, Chinese lizard genus measured six kinds of mitochondrial gene sequences, binding NCBI database already reported Eremias argus and Eremias brenchleyi mitochondrial genome sequence, between China investigate the case of lizard species 8 the phylogenetic position to provide molecular evidence to solve the case of lizard phylogenetic relationships. The main conclusions are as follows:1. This measured lizard genus six mitochondrial genome sequences are as follows: Eremias vermiculata(19914bp), Eremias multiocellata(19385 bp), Eremias przewalskii(18225 bp), Eremias velox(18033 bp), Eremias arguta(16475 bp) and Eremias grammica(15238 bp). Among them, the mitochondrial genome of Eremias vermiculata, Eremias multiocellata and Eremias przewalskii has been fully determined, while Eremias velox, Eremias arguta and Eremias grammica determined in addition to the control region of mitochondrial genome. By analyzing the determined data, obtained six species of mitochondrial genome sequence and the NCBI database reported Eremias argus and Eremias brenchleyi of the order, exactly the same genetic make-up and direction of transcription, including a major non-coding control zone(D-loop control region), a light chain end replication origin(OL structure), two ribosomal genes(12S rRNA and 16 S rRNA), 13 protein-coding genes(ATP6, ATP8, COI-III, ND1-6, ND4 L, Cytb), 22 tRNA genes.2. Eremias vermiculata, Eremias multiocellata, Eremias przewalskii, Eremias velox, Eremias arguta and Eremias grammica mitochondrial genes have AT biased,they are A + T content: 59.8%, 60.1%, 58.2%, 58.0%, 58.0%, 57.8%.3. Eremias of protein-coding genes, ATA is the most used initiation codon, ATGfollowed; TAA is the most used stop codon.4. Measured six kinds of lizard species of tRNA genes, can be folded into the typical cloverleaf secondary structures. Among them, there are generally a base mismatches imagine, most of the mismatches U ? G mismatch between bases.5. Phylogenetic relationship display, Eremias in China are a monophyletic group,genetic distance of Eremias multiocellata and Eremias przewalskii is particularly small, Eremias argus and Eremias brenchleyi form sibling species.
