Opisthopappus Shih,belonging to Chrysantheminae of Asteraceae,has two tentative species: Opisthopappus taihangensis(Ling)Shih and Opisthopappus longilobus Shih.The living environment of Opisthopappus Shih is special,and only distributed in the crevices of rocks in high altitude areas.It is of great significance for the scientific protection and development of Opisthopappus Shih to analyze the survival mechanism of its special habitat.In this study,the rhizosphere soil and roots of geographical locations of O.taihangensis(Hongdouxia,Jingnianghu,Gaojiatai,Xingtaidaxiagu,Hezhangxiang)and O.longilobus(Guanshan,Qinglongxia,Linshizuting,Wanxianshan,Dashuangcun)were studied by amplifying the Gene v3-v4 of bacterial 16 Sr RNA and fungal ITS,using High-throughput Hiseq sequencing technology and bio-informatics for species composition and difference analysis.The results showed that there were 534 rhizosphere bacterial,334 rhizosphere fungus,495 endophytes bacterial and 483 endophytes fungus for O.taihangensis and O.longilobus;14 rhizosphere bacterial?45 rhizosphere fungus?52 endophytes bacterial and 8endophytes fungus for O.taihangensis,14 rhizosphere bacterial?33 rhizosphere fungus?26 endophytes bacterial and 7 endophytes fungus for O.longilobus.There are significant differences at P<0.05 in the richness of 9 rhizosphere microorganism and 4endophytes microorganism.There are extremely significant at P<0.01 differences in the richness of 33 rhizosphere microorganism and 41 endophytes microorganism.Proteobacteria is the dominant bacteria among rhizosphere microorganisms and endophytes for O.longilobus,while Ascomycota is the dominant fungi.The results of UPGMA analysis showed that there was a high diversity of rhizosphere microorganisms between O.taihangensis and O.longilobus,and the geographical location was the key factor affecting the composition of rhizosphere microbial community.RDA/CCA analysis showed that the influence degree of climate factors on different samples was different,and the influence degree of rhizosphere microorganisms and endophytes dominant bacteria on different samples was also different.By microbiological and molecular biological techniques,rhizosphere microorganism and endophytes were isolated and purified,4 bacteria and 5 fungi degradation of cellulose,8 bacteria and 2 fungi phosphorus solubilizing strains and5 bacteria and 5 fungi potassium solubilizing strains were screened OTU.Pot experiments showed that the fungus QLX23 increased the height,fresh weight,dry weight and chlorophyll content of Arabidopsis thaliana significantly and it was identified as Trichoderma harzianum.In this experiment,the species,richness,function and diversity of the rhizosphere microorganisms and endophytes of Opisthopappus Shih were preliminarily defined,which provided important theoretical and technical guidance for the analysis of the special habitat ecological adaptation mechanism and adaptive evolution of Opisthopappus Shih.
