The idea of molecular computer was first put forward by Richard Phillips Feynman,the most famous physicist of America.However,at the time of the technical conditions the idea of”super microcomputer" is too difficult to achieve.The university of southern California,Professor Leonard Adleman used oligonucleotides to solve the classical graph theoretical problems in mathematics—Directed Hamiltonian Cycle Problem,experimently justifying the feasibility of molecular computer in 1994.Because of the ability of large-scale parallel computing and the capacity of efficient data storage,molecular computing becomes one of the most important ways to develop the high performance computer.Biological computing received extensive attention of researchers in recent years,more and more scientists devote to the study of molecular computer.In this paper,we summarized the research achievements of domestic and foreign scholars in the field of molecular computing and the use in this field of the related technology and materials.Then,we have designed two logical systems on the basis of previous work in our team.The work of this paper is as follows:(1)Firstly,this paper introduced the background and significance of DNA computing,and then we summarized the researches in this field on at home and and the developmental trend of biological computing.(2)Then we introduced the molecular computing technology and its relating researches.And this paper emphatically introduced two kinds of technologies:the DNA strand displacement and the fluorescence labeling technique.We introduced several kinds of nanomaterials which are widely used in biological computing,including ions,deoxidization ribozymes,G-quadruplex and so on.(3)In chapter 3,this paper built two metal-mediated logic models based on the principle that DNA matching can be mediated by metal ions.Then,we designed a cascaded logic model based on the two metal-mediated logic models.In this model,the metal ions are inputs and the output is the change of fluorescence intensity.(4)Finally,we introduced two biochemical molecules,streptavidin and DNAzyme.At first,we built a double inputs AND gate which could realize the process of the fluorescent signal weakens.Furthermore,we designed a multi-input AND gate with the two subunit of DNAzyme as input respectively.This gate implements the progress of fluorescence intensity increases.To increase the application value and the generality of this model,we designed a small molecular switch with both logic gates together.
