| Objective:To construct a rat model of polycystic ovary syndrome-insulin resistance(PCOS-IR)with trazozole and high-fat diet,and explore the effect of Yishen Quzhuo Formula on PCOS-IR model Effects of rat weight,abdominal circumference,ovarian volume,and number of follicles in the ovary,and to study the effects of prescriptions on androgen receptor(AR),luteinizing hormone receptor(LHR),Insulin receptor(INSR),insulin-like growth factors 1 receptor(IGF-1R),insulin receptor substrate 1 receptor(IRS-1R)The effect of several receptors on the expression of ovaries,stomach base,colon,and pituitary.Based on this,the therapeutic effect and mechanism of Yishen Quzhuo Recipe on PCOS-IR were explored.Methods:A total of 108 female SPF-grade SD female rats aged 6 to 8 weeks were randomly divided into 9 groups,which were divided into blank group,model group,Yishen Quzhuo high,medium and low dose group,Yishen group,metformin group,and Daying-Thirty-five groups,metformin combined with Dain 35 groups,12 in each group.The blank group was given ordinary feed,and the rest of the groups were given letrozole gavage combined with high-fat diet until the 27th day,and then the corresponding drug was given gavage for 2 weeks.During the feeding period,the rats' weight,length,and abdominal circumference were measured regularly.And calculate Lee's index of rat(an index to evaluate the degree of obesity in adult rats).After the treatment,the ovary,pituitary,bottom of the stomach,and colon were taken.The rat ovary volume and weight were measured,and the ovarian index was calculated.The tissues were dehydrated to make paraffin sections,and the ovaries were HE stained,and the number of follicles in the ovaries was calculated under a light microscope.Immunofluorescence was used to detect the expression of AR,LHR,INSR,IGF-1R,IRS-1R in the ovary of each group of rats;the expression of AR,LHR,INSR,IGF-1R in the bottom of the stomach and colon of the rats was detected;The pituitary AR,LHR,and IGF-1R expressions were detected.Results:After 14 days of treatment,it was found that the weight,abdominal circumference,and Lee's index of the rats in the Yishen Quzhuo recipe group were lower than those in the model group(P<0.05),and the ovarian volume and ovarian index were reduced(P<0.05).Compared with the group,abnormally increased follicles decreased(P<0.05).In the Yishen Quzhuo recipe group,the IOD values of AR,LHR,and INSR expressions decreased(P<0.05),IGF-1R expressions increased(P<0.05),and IRS-1R did not change significantly(P>0.05).The expression of AR in the bottom of the stomach decreased(P<0.01),the expression of LHR and INSR in the stomach and intestines decreased(P<0.05),and the expression of IGF-1R increased(P<0.05).There was no difference in the expression of AR in the colon(P>0.05).The expression of AR and LHR on pituitary decreased(P<0.05),and the expression of IGF-1R increased(P<0.05).Conclusion:Yishen Quzhuo Fang has good therapeutic effect on PCOS-IR,and the overall effect is better than that of Yishen Traditional Chinese Medicine group.Its mechanism of action may be to improve the environment of high androgen and high insulin of the ovary by affecting the hypothalamic-pituitary-ovarian axis;and to improve the expression of several receptors in the gastrointestinal tract,promote the normalization of gastrointestinal metabolism,and connect the stomach The effect of the intestine on the reproductive axis,blocks the vicious cycle between endocrine and metabolic disorders,and achieves the purpose of restoring the normal ovulation function of the ovary and treating diseases. |
