Purpose:Tourette syndrome(TS)is also known as Tourette syndrome.The clinical features of this disease are rapid and irregular twitching of facial muscles,neck muscles,or limb muscles,and/or strange noises in the larynx,and even profanity.In recent years,the incidence of TS is increasing.Foreign studies have shown that the incidence of TS is about 0.3%to 0.9%,and the clinical symptoms of children may always be present or worse.The etiology of TS is complex and its manifestations are diverse.Its etiology and specific mechanism are still under debate.This experiment will explore the relationship between TS pathogenesis and cellular immunity by comparing the levels of T-lymphocyte subsets between rats in the blank group and IDPN-induced TS models.By analyzing the changes in various indexes of T lymphocyte subsets before and after treatment of TS model rats by Jianpi Zhidong Decoction,the immunological mechanism of Jianpi Zhidong Decoction in the treatment of this disease was explored to provide Jianpi Zhidong Decoction for the treatment of TS Theoretical basis.Combining the results of this experiment and the literature on the pathogenesis of TS,we will continue to explore the possible mechanisms of its pathogenesis from the perspective of cellular immunity.Method:36 male SD rats,weighing 50g±10g,were randomly divided into 4 groups according to body weight using a random coding table,namely blank group,model group,western medicine group,and traditional Chinese medicine group,with 9 rats in each group.Except for the blank group,rats in the remaining three groups were intraperitoneally injected with IDPN 250 mg/kg·d,and the blank group was intraperitoneally injected with an equal volume of saline once daily for 7 consecutive days.After successful modeling,the blank group and the model group were gavaged with 0.9%Nacl solution(10ml/kg·d),the western medicine group was gavaged with Tiberium suspension(21ml/kg·d),and the Jianpizhidong decoction group The Chinese medicine granule suspension(16ml/kg·d)was intragastrically administered for 4 weeks,and the behavioral tests were performed on the first day after modeling and on the 8,15,22,and 29 days after administration.Blood was taken from the abdominal aorta 24 hours after the last administration,and the CD3+,CD4+,CD8+ and CD4+/CD8+ values in serum were determined by ELISA.Result:1.Compared with the model group,Jianpi Zhidong Decoction can reduce the stereotype exercise score of TS model rats,the difference is statistically significant(P<0.05)2 In this experiment,there was no significant difference in CD3+,CD4+,CD8+ and CD4+/CD8+ between the blank group and the model group(P>0.05).3 Compared with the model group,the CD3+ of the Chinese medicine group and the Western medicine group decreased,which was statistically significant(P<0.05).4 Compared with the model group and the Western medicine group,the CD4+ and CD4+/CD8+ of the Chinese medicine group increased,which was statistically significant(P<0.05).Conclusion:1.Jianpi Zhidong Decoction can significantly reduce the stereotyped movement of TS rats.2.In the early stage of the research group,IDPN-induced TS rats were used to conduct a number of experiments and research on the pathogenesis.However,in this experiment,the IDPN-induced TS rat model showed no significant differences in T lymphocyte subsets.The following three Possible possibilities:The change of T lymphocyte subsets in TS rats is really not obvious compared with the blank group.You can choose other immune factors for further research;the second is to consider the small sample size and affect the results;the third may be related to IDPN It is related to neurotoxin drugs.Although it can better change the behavioral changes and neurotransmitters of the model,it fails to reflect the immune disorders that TS may cause.It is suggested that when conducting related immunological studies of TS,it is necessary to consider and tap other The model of validity and structural validity,using the serum of TS patients for modeling,may be closer to the immune pathogenesis of TS,reflecting structural validity.3.Compared with the model group,the levels of CD3+ in the western medicine group and the traditional Chinese medicine group decreased.The consideration is related to the fact that this experimental model can only reflect its surface validity and fails to reflect the structural validity of the TS rat's immunity,and it cannot truly reflect the effect of Jianpi Zhidong Decoction on CD3+ levels.It still needs to be in future experiments.research.4.Compared with the model group and the western medicine group,there is a significant difference in the increase of CD4+ and CD4+/CD8+ in the traditional Chinese medicine group,suggesting that Jianpi Zhidong Decoction increased the level of cellular immunity in the treatment of TS model rats.However,whether improving CD4+ and CD4+/CD8+ is the therapeutic target of Jianpi Zhidong Decoction in treating children with TS remains to be confirmed.
