Research objective Tourette syndrome(also known as Tourette's,TS,Gilles de la Tourette syndrome or GTS),is a neuropsychiatric disorder characterized by multiple motor tics associated with vocal tics and language and behavior disorder.In Chinese medicine,according to its typical clinical manifestations,the condition can be classified under chronic infantile convulsion or Chi Zong,liver-wind.Its causes are complex and not yet clear.Nowadays,most physicians think TS has various causes related to genetic,neurotransmitter imbalance,psychological factors,etc[1].The disease usually beginning in childhood and adolescence,and boys obviously exceed girls.with the increase of pollution and stress factors,the incidence of the disease in recent years gradually increased[2],and should be further valued highly in society.As the etiology and neuro-chemical basis remains unknown,there is still no specific or effective treatment or cure in modern medicine and easy to repeated.So it is difficult to obtain ideal clinical effect only by a single approach.We should focus on the traditional Chinese medicine and take comprehensive treatment of the disease.Professor Wang su-mei has been committed to the research on treatment of Tourette syndrome and its etiology.TS is characterized by complex symptoms,irregular change,poor prognosis and easy to repeated.In Chinese medicine,it can be attributed to "strange disease".Professor Wang summarized her many years clinical experience,think that the disease is asthenia in origin and asthenia in superficiality.Wind and phlegm are key factors.The pathological location mainly involves the liver and spleen.Her self-made decoction is based on the principle of supportting earth to inhibit wood.And there made some progress based on previous experimental and clinical research.Combined with the neurotransmitter imbalance theory,the purpose is to study the mechanism of Jian pi zhi dong decoction,and the target in the intervention of treatment with TS.This study based on the early stage of the series of experiments,use animal behavior evaluation and Real-time PCR method,to test the stereotyped behavior of IDPN result in TS model rats,autonomous activity,and the brain striatum glutamate transporter EAAT2 content,in order to further reveal the mechanism of action of Jianpizhidong decoction.This experiment is divided into four parts:(1)to compare and observe the TS model rats in stereotyped behavior score changes before and after the treatment(2)to compare and observe the TS model rats in the autonomous activity of distance change before and after the treatment(3)the application of Real-time PCR technique to detect glutamate transporter EAAT2 expression level changes in the brain striatum in TS model rats before and after treatment.The main results were as follows:Methods40 male SD rats,weight(200g± 10g).Divided into 5 groups according to the random coding table,which are blank group,model group,Tiapride(Tia)group,and JPZD group of middle dose and high dose.Each group contains eight and were injected with IDPN(250 mg,kg-1,d-1.)once a day for 7 consecutive days.The blank and model groups were gavaged with saline(0.9%)at 10 ml*kg-1,the group with Tia at 21mg-kg-1,the group with JPZDD of middle dose at 16g.kg-1,and the group with JPZDD of high dose at 32g.kg-1 respectively once a day for 8 consecutive weeks.Every 7 days double blind method was carried out on the model rats rigid sports scores,and independent activity determination.After the last observation,use RT-PCR technology to test the excitatory amino acid of glutamate transporter EAAT2 in the brain striatum.ResultsThe stereotyped behavior in rats and independent activity score are steadily falling after treatment of JPZDD(2):Compared with blank control group,the expression of EAAT2 in model group increased significantly(p<0.01);the expression of EAAT2 in the group with Tia,JPZDD of middle and high dose are significantly lower compared to model group(p<0.01),but no statistical significance between the three.ConclusionsJPZDD have a significant inhibitory effect to stop the stereotyped behavior and automatic activities of TS rats,its targets may be linked to the concentration of excitatory amino acid transporter EAAT2 in the brain striatum.
