Objective:Yujin injection is made up of h.cordata and honeysuckle extracts of TCM,curative effect,clinical commonly used in the treatment of acute upper respiratory tract infection related diseases.However,there are few studies on the new way of atomizatied inhalation of yujin injection.Therefore,this project through the animal experiment to research whether Yu jin injection atomization inhalation has antipyretic,anti-inflammatory,antitussive,expectorant,pharmacodynamics function and the inhibition of some common pathogens in vitro,so as to illustrate the antibacterial activity of yujin injection in vitro and the pharmacodynamics of atomization inhalation,provide experimental basis for the application of the clinical.Methods:1.bacteriostatic experiment in vitro:microbroth dilution methods were carried out to observe the Minimum inhibitory concentration(MIC)of yujin injection on 13 tested strains,including Staphylococcus aureus standard strain ATCC29213 and clinical isolates,Escherichia coli standard strain ATCC25922 and clinical isolates,Streptococcus pneumoniae standard strain ATCC49619 and clinical isolates,Group A-?hemolyticstreptococcus standard strain ATCC32204,Pseudomonas aeruginosa standard strain ATCC27853 and clinical isolates and slime type,Klebsiella pneumoniae standard strain ATCC700603 and clinical isolates and slime type.2.Pharmacodynamics experiment:(1)the antipyretic experiment:Mice fever model evoked by 15%yeast were used to observe the effect of atomized inhalation of Yujin injection on body temperature,the levels of Interleukin-6(IL-6),tumor necrosis factor(TNF-?),interleukin-1?(IL-1?)in the serum,and the levels of and prostaglandin E2(PGE2)in the hypothalamic Of mice.(2)anti-inflammatory test:the models of mice auricle swelling caused by xylene were used to observe the inhibition rate of atomized inhalation of yujin injection on the ear swelling of mice.(3)antitussive test:the effects of of atomized inhalation of yujin injection on the number of coughs and the incubation period of coughs,as well as the contents of interleukin-10(il-10)in the serum,were observed in mice induced by ammonia solution.(4)expectorant test:phenol red excretion method was used in the trachea of mice to observe the effect of atomized inhalation of yu jin injection on phenol red excretion.Results:1.Yu jin injection had a certain inhibitory effect on some bacterias that could induce acute respiratory tract infection:(1)the MIC value of the standard strain ATCC27853 of pseudomonas aeruginosa with bacterial concentration of 104 was 2.5 ?g/ml.(2)the MIC values of standard strains of streptococcus pneumoniae ATCC 49619 and clinical isolates with bacterial suspension concentrations of 104 and 105 were 0.039 ?g/ml,0.15625 ?g/ml,0.3125 ?g/ml and 1.25 ?g/ml,respectively.(3)the MIC values of ATCC 32204,the standard strain of Group A-?hemolyticstreptococcus with bacterial suspension concentrations of 104 and 105,were 0.15625?g/ml and 0.3125 ?g/ml,respectively.2.Yujin injection atomization inhalation of antipyretic,anti-inflammatory,cough,eliminating phlegm,pharmacodynamics experiment research shows that:(1)the Yujin injection high-dose group of atomizing inhalation of dry yeast to rat fever model has significant antipyretic effect,(2)the Yujin injection medium-dose group of atomizing inhalation has significant inhibitory effect on mouse ear swelling induced by xylene.(3)the high-dose and medium-dose groups of Yujin injection could significantly reduce the number of coughs,and the high-dose group could prolong the incubation period of mice induced by the stimulation of concentrated ammonia water,considering that the possible mechanism of action was to increase the production of il-10 in the serum of the coughing mice.Conclusion:Yujin injection has a significant inhibitory effect on some bacteria which can induce acute upper respiratory tract infection in vitro.And the atomizatied inhalation has obvious antipyretic,antitussive and anti-inflammatory effects.
