Objective:The study that the different concentrations of potassium chloride after atomization inhalation at different serum and bronchoalveolar lavage fluid of TNF-a, IL-6, CRP expression level, lung coefficient, the pathologic lung tissue pathology morphology change, for the safety of potassium chloride atomization inhalation provide theoretical basis for potassium chloride, atomization inhalation therapy hypokalaemia provide the feasibility of the evidence. Methods:After feeding3days guinea pigs repurchase adaptability in the future according to the suction potassium concentration will be different experimental animals computer random (by the statistics personnel produces random sequence) into A (ck) and B (5%concentration), C (10%concentration), D (20%concentration) group. Blank control group only5each group, each group of the15only, a total of only fifty, blank control group do not intervention, intervention will B, C, D three groups of experimental animals were in50cm×50cm×50cm not completely airtight glass box, atomization inhalation potassium chloride and20min/time, interval12h, respectively on1D,3D,7D each group extraction5different animals to return biochemical and lung tissue samples. The last time aerosolized potassium chloride, according to300mg/kg dose intraperitoneal injection of10%chloral hydrate anesthesia, take apex blood5ml,4℃water mixture standing and min,4℃low temperature centrifugal (1800r,20min), separate serum biochemical test. In aseptic condition descending neck midline incision gas separation, ligation on the right side of the main bronchus to left lung, in37℃preheat0.9%NS into2ml repeated lavage3times, light pressure guinea pig lung collecting bronchoalveolar lavage fluid, recovery is more than70%(1800r centrifugal and min), collecting bronchoalveolar lavage supernatant fluid,-80℃frozen storage. Bronchoalveolar lavage finished, beheaded put to death animals, the chest take the upper leaves, in10%of year’s marin solution soak fixed, and h after alcohol in turn dehydration, xylene transparent paraffin embedding,4um serial section, with wood grain-eosin staining, light-microscopy lung tissue morphology change. Take the right lower lobe organization remove redundant connective tissue, filter paper dry lung surface liquid with electronic balance precision weighing is the wet weight, the specimen in80℃temperature drying oven72h to weight constant, again say its weight as dry weight, calculation lung wet, dry weight ratio, lung coefficient calculation.Results:1,20%concentration group two time of1d,3d TNF-a, IL-6in bronchoalveolar lavage fluid expression level and blank control group more statistical significance,7d the same point the results of variance analysis indifference. Four groups guinea pig bronchoalveolar lavage fluid at the level of CRP were no statistical significance, P>0.05;2, four groups of guinea pig serum TNF-a, IL-6, CRP expression and lung tissue than wet dry weight of each time the results of variance analysis were no statistical significance, P>0.05;3, four groups of guinea pig pathological results show that:the blank control group,5%concentration group,10%concentration group after atomization inhalation potassium chloride lung tissue paraffin section alveolar structure is clear, the alveolar walls thin, alveolar walls and interstitial cells without inflammatory infiltration,20%concentration group have lung tissue edema, alveolar cavity and interstitial have a small amount of red blood cells and inflammatory cells infiltration, alveolar complete structure.Conclusion:1, Clinical dosage form (<10%) potassium chloride atomization inhalation is safe that I can recommend;20%of potassium chloride concentration atomization inhalation not recommended for routine;...
