Purpose: Knee,Osteoarthritis(KOA)is a clinically common orthopedic disease.The main pathological manifestations are cystic changes of articular cartilage,destruction of articular cartilage,osteochondral necrosis of the articular cartilage,and a non-specific inflammation formed around the joints and even the subchondral bone hyperplasia.Mainly in the knee joint stiffness,pain,dysfunction and even joint deformation and other symptoms for clinical manifestations,severe cases can be disabled,the aging of China's social population is becoming more serious,the number of patients with osteoarthritis has increased significantly,people over 60 years old Among them,50% of the population had KOA performance in X-ray,35%-50% had clinical manifestations,early symptoms of KOA were not obvious,a few patients had painful performance after excessive activity,and the condition improved after rest,X-ray abnormality Less change,only MRI can be seen in mild cartilage damage [1],most patients mistakenly believe that it is caused by fatigue,did not promptly interfere with the treatment,so in the treatment of KOA should be early diagnosis,early treatment,in the patient's joints The cartilage has not yet developed lesions,the prosthetic space has not been narrowed before the prophylactic treatment and comprehensive treatment,the early experimental study of rabbit knee osteoarthritis by the method of phlegm and blood stasis,observe the value of Huayu Decoction in the treatment of early osteoarthritis The mechanism of action provides pharmacological evidence for its clinical application.MATERIALS AND METHODS: Twenty-eight rabbits provided by the Experimental Center of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine were modeled by modified Hult method and then randomly divided into 4 groups,each group consisting of 7 groups.The groups were normal group and model group.After the successful model establishment,the normal group and the model group were given 2 ml/d of distilled water,and Qutan Huayu group was administered with 3.85 g/d/kg of crude drug dissolved in 5 ml of normal saline.The Futalin group was dissolved in 5 ml of distilled water at a dose of 10.4 mg/d/kg.The rats were orally administered for 4 weeks.The animals were sacrificed after the blood was collected from the ear artery.The rabbit knee joint cartilage andsynovium were taken out,and the articular cartilage specimens were observed and lighted.Microscopic analysis and ELISA were used to detect the levels of IL-1 and TNF-? in serum and joint fluid.Results:1.After observing the surface of cartilage tissue of rabbit knee,the OARSI score of model group was higher than that of normal group,P <0.01,which was statistically significant.The OARSI scores of the Qutan Huayu group and the Futalin group were lower than those of the model group,P <0.05,which were statistically significant.2.The Mankin 's score of the HE staining model group was higher than the normal group,P<0.01,which was statistically significant.The Mankin 's score of the Qutan Huayu group was lower than that of the model group,P <0.05,which was statistically significant.The Mankin 's score of the Futalin group was lower than that of the model group,P> 0.05,which was not statistically significant.3.The levels of IL-1 and TNF-? in the serum and synovium of the model group increased compared with the normal group,which proved that the modified Hulth method was successfully used for modeling.The content of IL-1 and TNF-? in serum and synovium of expectorant stasis group was lower than that of model group,P <0.05,which was statistically significant.The content of IL-1 and TNF-? in serum and synovium of the futalin group was lower than that of the model group,P <0.05,which was statistically significant.Conclusion:1.This experiment successfully applied the modified Hulth method to induce rabbit knee KOA model.Observation of OARSI scores showed that Qutan Huayu Decoction could improve rabbit knee arthritis to a certain extent and have protective effect on articular cartilage.2.Mankin's score of HE staining indicated that Qutan Huayu Decoction could protect the articular cartilage of rabbits with knee osteoarthritis.3.Qutan Huayu Decoction have a reducing effect on IL-1 and TNF-?in serum and synovium,inhibit the inflammatory response of articular cartilage synovium,promote the recovery of tissue morphology,and thus have an intervention effect on early KOA...
