| Objective: To observe the effect of Huoxue Huayu Decoction made by Chinese angelica,radix paeoniae rubra,by Jiang Huang of IL-1,the content of IL-6 of rabbit knee joint fluid,the curative effect of Huoxue Huayu method in treatment of rabbit knee osteoarthritis,and provide the experimental basis for promoting blood circulat tion and removing blood stasis clinical treatment of knee osteoarthritis.Methods: a total of 42 New Zealand rabbits were randomly divided into two groups,with a total of 30 rats in each group.The model group was composed of a total of only 3 rats and sham operated group(n = 6).The modeling method according to the modified Hulth model model,after 4weeks,the model group of 3 randomly selected only according to X-ray,determine the success of modeling by X-ray,afterinjecting and then the rats were randomly divided into the TCM group,positive drug group,10 rats in each group.The traditional Chinese medicine group was randomly divided into 2 groups,namely,the traditional C hinese medicine high concentration group and the normal concentrattion group,and the drug concentration in the high concentration group was about 3 times higher than that in the normal group.The traditional Chinese medicine group was treated with radix paeoniae rubra,Angelica sinensis and Jiang Huang mixture,and the positive drug group was given glucosamine hydrochloride orally.The sham oper ation group and the normal group were given the same dose of normal saline.After four weeks,4-6 rabbits were taken forradiograph ic knee joint cartilage slice observation,and test the content of synovial fluid in IL-1,IL-6.Results: 1 groups of rabbit knee radiographs before and after model ming:compared with sham operation group,normal control group,model group after 4 weeks of joint bone hyperplasia,joint space narrowing,unequal width,that arthritis rabbit model has been establ ished suces sfully.2observation of histomorphology:model group(model 8 weeks and after 4 weeks of administration)edge defect,not smooth growth is not the rule,partof the cartilage necrosis,cartilage mat rix and dissolution,synovial hyperpla sia,cartilage surface cells thinnig,cartilage cell shedding into bone marrow;traditional Chinese medicine ine group is not the law of growth,the disorder,synovial thickening and thickening,r ough articular surface,no cartilage necrosis,relative to model grou p is better;the traditional Chinese medicine group,positive drug co ntrol group after 4weeks,IL-1 and IL-6 in synovial fluid were lower than the model group(P<0.05)traditional Chinese medicine group an d positive drug control group IL-1 content in synovial fluid between the groups by gavage after 4weeks there was no significant differenc e(P>0.05),the differences of thecontent of IL-6(P < 0.05),Chinese medi cine group is better than western medicine group;after 4 weeks,the sham operation group and normal group without significant differe nces between(P>0.05),3 rabbits each group Markin score: the blank g roup: 3.40±0.85 points;sham operation group s:3.43±0.40 points;model group: 9.56±0.73 points;Chinese medicine group: 6.5 4±0.55points;pos itive drug control group:6.16±0.78.The traditional Chinese medic ine group and positive drug control group scores were lower than the mode l group t here weresignificant differences between the groups(P < 0.05),no significant differ ence between the control group and the sham group(P>0.05);traditional Chinese medicine group and positive drug control group,no significant difference between groups(P>0.05).Conclusion:The mixture of Radix Paeoniae Rubra,Jiang Huang and Angelica can effectively reduce the content of interleukin in the joint fluid of rabbit model,especi ially the content of IL-6.There was no significant difference between the experimental data and the posti ive drug control group,which showed that the method ofpromoting bloo d circulation and removing blood stasis could play an important role in preventi ng and treating knee osteoarthritis. |
