Purpose:In this experiment,the rat model of acute myocardial infarction was induced by high-fat feeding combined with isoproterenol hydrochloride.The effect of acupuncture therapy combined with qijingtongmai Decoction on the expression of transient outward potassium channel related protein Kv4.3 and auxiliary protein kchaip2 in the myocardial cells of acute myocardial infarction rats was studied.At the level of ion channel,the therapeutic effect of acupuncture combined with qijingtongmai Decoction on acute heart was explored The possible mechanism of myocardial infarction and its advantages were verified.Material and method:65 healthy SPF male SD rats with body weight of(200 ± 20g)were randomly selected as the blank control group according to the random number table,and the rest 55 rats were fed with high-fat diet for 3 weeks(high-fat group).65 rats were anesthetized with 10% chloral hydrate by intraperitoneal injection in the left lower abdomen.Using bl-420 s biological function experiment system,the ECG of lead Ⅱ in normal condition of mice was recorded.The rats in the blank control group were injected with normal saline(85mg / kg)subcutaneously at the root of limbs at one time,and the other 55 rats in the high-fat group were injected with isoprenaline hydrochloride(85mg / kg)in the same way to prepare the animal model.All rats were injected for the second time at an interval of 24 hours,and then the ECG was recorded again.According to the ECG changes before and after modeling,40 rats were randomly divided into model group,acupuncture combined with medicine group,acupuncture group and Western medicine group,10 rats in each group.15 days after the success of the model,the artificial intervention was carried out.The blank group and the model group were given normal saline at 8:00 and 18:00 respectively.The combination group was given qiaotongmai Decoction at 8:00 and 18:00 respectively.At 10:00every day,Huatuo brand acupuncture needle(0.18 mm × 25mm)was used to prick into the subcutaneous skin of each acupoint of the rats for about 2mm.After the rats were stable,the electric acupuncture instrument was turned on and the density wave was used with the frequency of 2-20 hz.The current intensity was as follows: slight tremor occurred at the acupuncture acupoint It is advisable to keep the needle for 15 minutes.The rats in the acupuncture group were treated with the above acupuncture method once a day for 15 days;the rats in the western medicine group were treated with aspirin and isosorbide mononitrate by gavage at 8:00 and 18:00 every day.During the treatment,the general living conditions of rats were recorded.After treatment,the ECG of rats was labeled.The expression of hs CRP in serum of rats was detected by ELISA.The expression of Kv4.3 and KCh IP2 in myocardial cells of rats was detected by Western blot.Results:1.Compared with the control group,the T-wave voltage of ECG in the model group was significantly lower,with a statistically significant change(P < 0.05).Compared with the model group,the T-wave voltage of ECG in the acupuncture drug combination group,the acupuncture group and the western medicine group were all increased(P < 0.05),and the T-wave voltage of ECG in the acupuncture drug combination group was the most obvious rise,and there was no significant difference between the western medicine group and the acupuncture group(P > 0.05)2.Compared with the control group,the expression of hs CRP in the serum of the model group was significantly increased(P < 0.05).Compared with the model group,the expression of hs CRP in the serum of the acupuncture drug combination group,the acupuncture group and the western medicine group was significantly reduced(P < 0.05),in which the expression of hs CRP in the acupuncture drug combination group was the most decreased,while the expression of hs CRP in the serum of the western medicine group was not significantly changed compared with the acupuncture group(P < 0.05)3.Compared with the control group,the protein expression of Kv4.3 and KCh IP2 in the model group was decreased(P < 0.05).Compared with the model group,the protein expression of Kv4.3 and KCh IP2 in myocardial cells of rats in the acupuncture combined group,acupuncture group and Western medicine group increased significantly(P < 0.05),especially in the acupuncture combined group.There was no significant difference between the western medicine group and the acupuncture group(P > 0.05)Conclusion:1.Acupuncture combined with qijingtongmai decoction has a good therapeutic effect on rats with acute myocardial infarction,which is mainly achieved by regulating the expression of potassium channel related proteins.2.The therapeutic effect of acupuncture combined with qijingtongmai Decoction on rats with myocardial infarction has obvious advantages over other treatment groups.
