Effects Of Acupuncture Combined With Xijingtongmai Decoction Pretreatment On Inward Rectifier Potassium Channel-related Proteins In Myocardial Infarction Rats

Purpose:The rats were pretreated with acupuncture and medicine,and then the rats were given acupuncture at Neiguan,Zusanli and Xinshu points,combined with the traditional Chinese medicine Xijingtongmai decoction.After the myocardial infarction model rats were reproduced,the pathological changes of myocardial tissue and Kir2.1,Kir2.2 and Kir2.3 of inward rectifier potassium channels were observed to provide in-depth data support and new ideas for TCM clinical prevention and reduction of myocardial infarction injury.Material and method:Fifty healthy male SD rats were randomly divided into 5 groups: blank group,model group,western medicine preconditioning group,traditional Chinese medicine preconditioning group,acupuncture combined with medicine preconditioning group,with 10 rats in each group.During this period,8 rats in each group were selected for ECG.The blank group and model group were given normal saline once a day for 14 consecutive days.The western medicine preconditioning group was given aspirin enteric solution once a day for 14 days.The traditional Chinese medicine preconditioning group was given the Xijingtongmai decoction once a day for 14 days.The acupuncture combined with traditional Chinese medicine preconditioning group was treated by gavage of the Xijingtongmai decoction combined with acupuncture at Neiguan,Zusanli and Xinshu points once a day for 14 consecutive days.Rats fed with high-fat diet for 3 weeks maintained the preconditioning pattern unchanged.The rat model of myocardial infarction was established by subcutaneous injection of isoproterenol(85 mg / kg / d)for 2 consecutive days.The ECG was continuously observed.When the st wave in lead II of ECG was significantly elevated,it indicated that the model was successfully established.The blank group was injected with the same amount of normal saline.After anesthesia,the tissues were quickly separated on the ice bag,the cardiac tissue was collected and the pathological changes of myocardial cells were observed.The data were analyzed by spss24.0 software,and the measurement data were analyzed by single sample analysis of variance,P < 0.05 was statistically significant.Results:1 The electrocardiogram(ECG)of the five groups of rats showed normal blunt round P wave and no S-T segment change during ordinary feed feeding(P>0.05);During the high fat diet,compared with the blank group,the S-T segment of the model group was elevated,and the T-wave was abnormal(P<0.05),the other three groups had no significant changes compared with those before high fat diet(P>0.05);After the injection of isoproterenol,compared with the blank group,the rats in the model group showed pathologic wide Q wave,significantly increased S-T segment arched back,and inverted T wave(P<0.05);The electrocardiogram of the acupuncture and traditional Chinese medicine preconditioning group,the Chinese traditional medicine preconditioning group and the western medicine preconditioning group showed T wave flattening and S-T segment elevation,but compared with the model group,the differences were statistically significant(P<0.05),while the S-T segment of ECG in the acupuncture and traditional Chinese medicine combination preconditioning group was the smallest.2 Compared with the blank group,the protein expression levels of Kir2.1,Kir2.2 and Kir2.3in the model group were significantly decreased(P<0.05),the protein changes of Kir2.1 in the traditional Chinese medicine pretreatment group and the western medicine pretreatment group were not statistically significant(P>0.05),while the expression level of Kir2.1 in the acupuncture combined with traditional Chinese medicine pretreatment group was increased,the difference was statistically significant(P<0.05);the difference was statistically significant(P<0.05).Compared with the model group,the protein expression levels of Kir2.1,Kir2.2and Kir2.3 in the western medicine pretreatment group and the acupuncture combined with traditional Chinese medicine pretreatment group were significantly increased(P<0.05);compared with the traditional Chinese medicine pretreatment group,the protein expression levels of Kir2.1,Kir2.2 and Kir2.3 in the western medicine pretreatment group and the acupuncture combined with traditional Chinese medicine pretreatment group were significantly increased(P<0.05),and acupuncture combined with traditional Chinese medicine pretreatment group increased more significantly(P < 0.05).3 The changes of pathological sections of the heart tissue of the five groups of rats were compared.The myocardial tissue of the blank group was arranged regularly and clear.In the model group,there were a large number of parallel collagen fibers,bundles and inflammatory cells were infiltrated.A large number of parallel or interlaced collagen fiber bundles,a large number of muscle fibers and nucleolysis were observed in the heart tissue of rats in the western medicine pretreatment group.The structure of myocardium in the traditional Chinese medicine preconditioning group was disordered and inflammatory cells were scattered at the edge of infarction.The number of residual cardiomyocytes in the acupuncture and traditional Chinese medicine combination preconditioning group were more than that in the model group,the western medicine preconditioning group and the traditional Chinese medicine group,the arrangement was orderly and regular,and the infiltration of inflammatory cells were less.Conclusion:1 The combination of acupuncture at Xinshu,Neiguan and Zusanli points and traditional Chinese medicine preconditioning can improve the ischemia state of cardiomyocytes caused by high fat feed.Compared with other groups,preconditioning with acupuncture at Xinshu,Neiguan and Zusanli points combined with Xijingtongmai decoction could significantly prevent and reduce the myocardial ischemia and infarction injury in isoproterenoline-induced myocardial infarction model rats.2 Acupuncture at Xinshu,Neiguan and Zusanli points combined with the preconditioning of Xijingtongmai decoction can improve myocardial ischemia and protect myocardial cells.3 Acupuncture at Xinshu,Neiguan and Zusanli points combined with the preconditioning of the Xijingtongmai decoction can protect the structure of cardiomyocytes,improve the ischemic state of cardiomyocytes and prevent the occurrence of myocardial infarction,reduce the infarction injury.
