Objective The experimental platform of acute myocardial infarction(AMI)rats was established to treat AMI rats by means of acupuncture and drug combination,simple acupuncture and single drug.The purpose of this study was to compare the efficacy of the three treatments and to explore the mechanism of the three treatments based on oxidative stress.Methods AMI rat model was prepared by ligating the left anterior descending branch of coronary artery.Fifteen healthy rats were selected as the blank group,and the sixty AMI rats successfully modeled were divided into 4 groups(model group,simple acupuncture group,single drug group,acupuncture and drug combination group),5groups in total,(1)Blank group: bondage;(2)Model group: ligation of left anterior descending coronary artery + bondage;(3)simple acupuncture group: ligation of left anterior descending coronary artery + bondage + acupuncture at both Neiguan and Xinshu points;(4)single drug group: ligation of left anterior descending coronary artery + bondage + injection of salvia miltiorrhiza polyphenolate;(5)acupuncture and drug combination group: ligation of left anterior descending coronary artery +bondage + acupuncture at both Neiguan and Xinshu points + injection of Salvia polyphenolate.Each group intervened once a day for 7 days.Echocardiography imaging after intervention on day 7.Observe the HE staining of myocardial tissue in each group;detect the concentration of CK-MB and c Tn T,MDA content and SOD activity in the serum of each group of rats;detect the content of salvianolic acid b in the myocardial tissue of single drug group and acupuncture and drug combination group.Results 1.Model preparation: 1 hour after ligation of the left anterior descending coronary artery,EF<55%,indicating successful model preparation.2.Ratechocardiography: after 7 days of intervention,EF values of blank group,single drug group and acupuncture and drug combination group were significantly higher than those of model group(P<0.05);EF values of simple acupuncture group were higher than that of model group,but there is no statistical significance(P>0.05);EF values of acupuncture and drug combination group was significantly higher than that of simple acupuncture group(P<0.05).EF values of acupuncture and drug combination group was higher than that of single drug group,but no statistical significance(P>0.05).3.The concentration of CK-MB and c Tn T in rat serum: After 7days of intervention,the concentration of CK-MB in the acupuncture and drug combination group was significantly lower than that in the model group(P<0.05);and the concentration of CK-MB in the simple acupuncture group and the single drug group was lower In the model group,but no statistical significance(P>0.05).The concentration of c Tn T in single drug group and acupuncture and drug combination group were significantly lower than that in model group(P<0.01);the concentration of c Tn T in simple acupuncture group was lower than that in model group,but there was no statistical significance(P>0.05);the concentration of c Tn T in acupuncture and drug combination group was significant lower than simple acupuncture group(P<0.01);the concentration of c Tn T in acupuncture and drug combination group was lower that of single drug group,but no statistical significance(P>0.05).4.HE staining of rat myocardial tissue: After 7 days of intervention,the model group showed large amounts of myocardial fibrillary coagulation necrosis and numerous neutrophils;simple acupuncture group showed a small amount of myocardial fiber coagulative necrosis and a small amount of neutrophils;a small amount of neutrophils can be seen in the single drug group,and myocardial cell hypertrophy;only a few neutrophils were seen in the acupuncture and drug combination group,and myocardial cells were slightly hypertrophic.5.MDA content in rat serum: After 7 days of intervention,the MDA content of blank group,simple acupuncture group,single drug group and acupuncture and drug combination group were significantly lower than that of model group(P<0.01);the MDA content of the acupuncture and drug combination group was lower than that of the simple acupuncture group and the single drug group,butthere was no statistical significance(P>0.05).6.SOD activity in rat serum: After 7days of intervention,the SOD activity of blank group,simple acupuncture group,single drug group and acupuncture and drug combination group were lower than model group,but there was no statistical significance(P>0.05).7.Concentration of salvianolic acid b in rat myocardial tissue: the concentration of salvianolic acid b in the acupuncture and drug combination group is higher than that in the single drug group,but there is no statistical significance(P>0.05).Conclusions 1.The therapeutic effect of the combination of acupuncture and medicine on AMI rats is better than that of the two alone.2.The mechanism of the combination of acupuncture and medicine is synergistic may be that acupuncture promotes the enrichment of drugs in the heart of AMI rats,synergistically enhances the body's ability to resist oxidation and clear membrane lipid peroxidation products,improves the body's oxidative stress state,and then improves Myocardial damage.
