Purpose : The aim of this experiment is to observe the effect of acupuncture points combined with self-made qijingtongmai Decoction on voltage-gated Kir6.1 in rats with myocardial infarction Objective to study the possible mechanism of acupuncture combined with xijingtongmai Decoction in improving myocardial infarction,and to provide experimental data basis for the research and development of new treatment methods for myocardial infarction.Material and method:Taking 65 healthy male SD rats as experimental samples,10 rats were randomly selected as the control group by contrast experiment.In the early preparation process,one-time multi-point injection of normal saline was carried out at the medial root of all rats' limbs.In order to ensure the accuracy of data,it is recommended to inject twice with an interval of 24 hours.In order to make a better experimental comparison,10 rats in the control group were fed with normal diet,and the other rats were fed with high-fat diet.At the same time,all the experimental rats were injected with isoproterenol(85mg / kg)at multiple points to complete the preparation of the model.Through the comparison of ECG before and after modeling,40 rats were randomly selected from the remaining 55 rats and divided into model group,western medicine group,acupuncture group and acupuncture medicine combination group.From the second day after successful modeling,rats were divided into groups for 15 days.According to the different groups,the rats were treated with different methods,the control group and model group were treated with normal saline injection,the western medicine group were treated with aspirin and isosorbide mononitrate solvent,the acupuncture group were treated with Neiguan point,Xinshu point and Zusanli point once a day;the acupuncture drug combination group were treated with qijingtongmai decoction,and the same acupoints as the acupuncture group were selected for acupuncture once.When acupuncture was applied,Huatuo brand acupuncture(0.18 mm * 25MM)was inserted into the subcutaneous area of the acupoints of rats for about2 mm,and electroacupuncture was added.The frequency modulation rate was 2-20 hz,the current intensity was 2m A,and the needle was kept for 20 min.The protein expression of ATP sensitive potassium channels(Kir6.1,Kir6.2)was detected by Western blot,and the changes of cardiac histomorphology were observed by HE staining light microscope and electron microscope.Results:1.Compared with the control group,there was a certain degree of ST segment elevation in the untreated hyperlipidemic rats,which was statistically significant.After treatment,compared with the model group,the western medicine group,the acupuncture group and the acupuncture medicine combination group all had changes in the experimental process,and the acupuncture medicine combination group had the biggest change,with significant difference.Although there are differences between western medicine group and acupuncture group to a certain extent,there is no practical significance due to the small difference.2.Compared with the control group,the protein expression of 40 experimental samples in the experimental group was increased,with significant differenceIt has experimental value.After treatment,compared with the model group,the protein expression levels of Kir6.1 and Kir6.2 in the western medicine group,acupuncture group and acupuncture drug combination group showed a downward trend,and the acupuncture drug combination group showed the most obvious decline,which has experimental value.The decrease degree of acupuncture group and Western medicine group was not significant,which had no experimental value.3.Compared with the control group,the model group without treatment had the largest necrotic area of myocardial cells;the acupuncture group and Western medicine group had less infarct area;the combination of acupuncture and medicine group had relatively clear morphology of myocardial cells and the least infarct area.Conclusion:1.Compared with the treated rats,the ST segment of ECG in the untreated rats showed a significant trend of elevation in myocardial infarction.After treatment,the ST segment of ECG showed a downward trend,especially in the acupuncture drug combination group,which proved that the effect of the acupuncture drug combination group was more obvious than other treatments.2.After treatment,the protein expression of Kir6.1 and Kir6.2 in western medicine group,acupuncture group and acupuncture drug combination group decreased,and the effect of acupuncture drug combination group was the most prominent.
