Purpose:In this study,puerarin,an effective component of pueraria lobata,a traditional Chinese medicine,was applied to NCI-H1299 cells of human non-small cell lung cancer to observe its effect on invasion and migration of NCI-H1299 cells and explore relevant molecular mechanisms.Material and method:1.The effects of different doses of puerarin on the proliferation activity of NCI-H1299 cells in vitro after 24,48 and 72 hours were determined by CCK-8 method,and the treatment time and drug dosage without obvious inhibition effect of puerarin on the proliferation activity of cells were selected as the subsequent experimental intervention conditions.Then the effects of puerarin on the migration ability and the effect on the cell invasion ability of NCI-H1299 cells of human non-small cell lung cancer were respectively detected by wound healing experiment and Transwell method.2.the expression of E-cadherin and Vimentin in NCI-H1299 cells was detected by Western blot.Results:1.According to the results of CCK-8 method,puerarin has a certain inhibitory effect on the proliferation activity of NCI-H1299 cells,which can be shown as dose-time dependence.The dose and time conditions for puerarin to have no obvious inhibitory effect on NCI-H1299 cells are puerarin with dose of 10,20 and 40?mol/L respectively,and the treatment time is24h;hours.The results of the cell wound healing experiment showed that compared with the negative control group,the wound healing area of the 10,20 and 40?mol/L puerarin drug group was significantly reduced,and the mobility of NCI-H1299 cells was significantly decreased with the increase of puerarin dose,with the 40?mol/L puerarin treatment group being the most obvious(p < 0.05).Transwell invasion test results showed that compared with the negative control group,the invasion rate of NCI-H1299 cells was inhibited by puerarin in a concentration-dependent manner,and the inhibition effect was the mostsignificant at 40?mol/L,the difference was statistically significant(p<0.05).2.the results of Western blot test showed that,compared with the negative control group,the E-cadherin protein expression in the cells showed a significant upward trend and the Vimentin protein expression showed a significant downward trend(p<0.05)with the increase of puerarin dosage after treatment in the 10,20,and 40?mol/L puerarin group.Conclusion:1.Puerarin can inhibit the proliferation activity of human non-small and fine lung cancer NCI-H1299 cells,and can effectively inhibit the migration and invasion of cells under the conditions of drug dosage and treatment time without obvious inhibition on the proliferation activity of cells.2.The possible mechanism of puerarin inhibiting invasion and migration of human non-small cell lung cancer NCI-H1299 cells is related to the increase of E-cadherin expression and the decrease of Vimentin expression in the cells,thus reversing the occurrence of epithelial-mesenchymal transition(EMT)of NCI-H1299 cells.
