Background Lung cancer is a major cause of cancer-related death around the world,high morbidity and mortality caused by the lack of effective treatment,killing about 1.5 million people a year.Lung cancer is divided into non-small cell lung cancer(non-small cell lung cancer,NSCLC)and small cell lung cancer,small cell lung cancer,SCLC),including NSCLC accounted for 85% of all lung cancer,about 75% of NSCLC patients have been found in the middle.Although the progress of basic and clinical research,but the overall 5-year survival rate of non-small cell lung cancer is only 15%.The occurrence and development of the NSCLC are influenced by many factors,performance for multiple stages of complex process,pathogenesis is not yet clear.Therefore,explore the mechanism of lung cancer,degradation and progress,looking for effective diagnosis and treatment of target is necessary.Extracted from plants and Chinese herbal medicine effective and side reaction of small components on the corresponding target has become a new way of research and development ofdomestic and foreign scholars antitumor drugs.Carvacrol chemical structure for5-isopropyl-2-methyl phenol,phenol is a single post,extracted from many plants,including fruits,vegetables,spices and herbs,carvacrol has many USES,such as the safety of additives used in cosmetics and food production process,thyme balm,aromatherapy essential oil is the main component.The pharmacological effects of carvacrol confirmed with multiple directions.Several studies show that the CV has anti-inflammatory antioxidant stress and anti-tumor and well as a variety of biological activities,and is much lower toxicity to normal cells.In recent years,the study found that carvacrol can inhibit the growth of a variety of tumor cells,such as liver cancer cell,metastatic breast cancer cell,such as CV for a wide variety of tumor cells can inhibit proliferation and induce apoptosis of CV resistance mechanism of lung cancer,however,has not yet been elucidated.The present study suggests that CV for non-small cell lung cancer A549 to promote the role of apoptosis,but the mechanism of action,there is no report.A549 and NCI-H1299 belong to non-small cell lung cancer cell line,but the NCI-H1299 genome DNA of P53 is missing,not expression of P53 protein,so the level of apoptosis and the mechanism of the two different,there are differences in the sensitivity and effectiveness of the anticancer treatment.CV for NCI-H1299 cell function,the influence of literature have not been reported at home and abroad.Based on this,we will have CV in non-small cell lung cancer NCI-H1299 cells,selection of cysteine aspartic acid proteolytic enzyme-9(caspase-9),matrix metalloproteinases 9(MMP 9)and its inhibitors(TIMP-1)as the targets,to observe the effects of CV effect on tumor cell apoptosis,and discussed the related mechanism,for the accurate treatment of non-small-cell lung cancer(NSCLC)provides a new strategy.Methods NCI-H1299 cells was treated with different doses of carvacrol(0.00?20.0?40.0?60.0?80.0 ?mol/L)for24 or 48 h,the viability of cells was evaluated by MTT assay;apoptosis was analyzed by flow cytometry(FCM);the effect of carvacrol on NCI-H1299 metastasis was analyzed by Transwell assay.The expression level of caspase-9?MMP-9 and TIMP-1were detected by Quantitative realtime-PCR and Western blot.Result After treatment with carvacrol,the viability of NCI-H1299 cells was suppressed dramatically(P < 0.05),neither in dose-dependent nor in time-dependent manner(P>0.05).After incubated with carvacrol for 24 h,FCM analysis indicated that Carvacrol effectively induces NCI-H1299 cells apoptosis(P < 0.05),but not dose-dependent(P > 0.05).The ability of invasion was decreased(P<0.05).Carvacrol inhibited the protein and mRNA expression levels of caspase-9,but increased the expression of MMP-9 and TIMP-1(P<0.05).Conclusion Carvacrol induces apoptosis of NCI-H1299 cells and inhibits their invasion,which maybe associated with via up-regulation of caspase-9expression and down-regulation of MMP-9 expression.
