Background: Lung cancer is the most common malignanciy.Its morbidity and mortality are the highest among malignant tumors.About 1.6 million people in the world are newly diagnosed with lung cancer every year.Although the treatment of lung cancer is increasing,the death rate of lung cancer remains high.Non-small cell lung cancer(NSCLC)is the main type of lung cancer,and its specific etiology and mechanism are still not clear.The tumor necrosis factor ?-inducible protein 2(TNFAIP2)has been proved to be an oncogene in many malignant tumors,such as nasopharyngeal carcinoma,breast cancer,and glioma,and it plays an important role in the occurrence and development of malignant tumors.The role of small cell lung cancer has not yet been reported.Therefore,we explored the expression of TNFAIP2 in non-small cell lung cancer,which is expected to provide a new way for the diagnosis and treatment of non-small cell lung cancer.Objective: To explore the expression of TNFAIP2(TNF-?-induced protein 2)in non-small cell lung cancer(NSCLC)and its effect on cell migration and invasion.Methods:(1)TNFAIP2 mRNA expression levels were examined by RT-PCR;TNFAIP2 protein expression levels were examined by immumohistochemical staining.(2)Lentivirus-mediated RNA interference(LV-TNFAIP2 mRNAi)of TNFAIP2 and Negative control lentivirus(LV-CON)were infected into H1299 cells.(3)The mRNA and protein expression of TNFAIP2 were tested by RT-PCR and Western blot,respectively.(4)The migration and invasion ability of cells was measured by Transwell assay.(5)CCK-8 were performed to analyze the cells proliferation.(6)Flow cytometry was used to detect the changes of apoptosis.(7)The changes of protein expression were detected by Western blot.Results:(1).The expression of TNFAIP2 in non-small cell lung cancer tissues was significantly higher than that in Paracancer(P<0.05).(2)Contrast to the negative control group,The expression of TNFAIP2 mRNA and protein of H1299 in LV-TNFAIP2 mRNAi was significantly lower(P<0.05).(3).The proliferation,migration and invasion ability of the cells decreased(P <0.05),the apoptosis rate increased(P <0.05).The expression of cell protein N-cadherin and Vimentin decreased,and the expression of E-cadherin increased.Conclusion: TNFAIP2 is associated with the occurrence and development of non-small cell lung cancer.Down-regulation of TNFAIP2 can inhibit cell migration and invasion ability through the regulation of EMT process.
