| Purpose:In this experiment,the content of vascular cell adhesion molecule VCAM-1 and interleukin IL-1β in rat serum after myocardial ischemia-reperfusion model intervention was measured by Shuangshen Tongmai Granules,then observe the morphological changes of rat cardiomyocytes under a light microscope.To investigate the effect of Shuangshen Tongmai Granules on myocardial cells in rats with ischemia-reperfusion injury and its possible mechanism,validate the effective role of Shuangshen Tongmai Granules in treating such cardiovascular diseases,and provide theoretical basis for clinical promotion of such traditional Chinese medicine treatments for diseases.Material and method:To prepare the Chinese medicine preparation Shuangshen Tongmai Granules in the hospital.Rats were randomly divided into six groups:control group,model group,western medicine group,traditional Chinese medicine low dose group,traditional Chinese medicine middle dose group and traditional Chinese medicine high dose group,ten rats in each group.The western medicine group was given hexinshuang,the Chinese medicine group was given different doses of shuangshentongmai granules,the drug was administered to the stomach once a day,and the other two groups were administered the same amount of saline for two consecutive weeks.Later,the left anterior descending coronary artery was ligated to create a rat model.The control group only performed thoracotomy and no ligation,thereby replicating a model of myocardial ischemia-reperfusion injury in rats.Serum was collected from the abdominal aorta of rats,and the contents of indicators in each group were determined by ELISA method according to the requirements of the kits of VCAM-1 and IL-1β,and myocardial tissue sections were made by HE staining.Results: 1.VCAM-1:Compared with the control group,the levels of VCAM-1 in the model group and each medication group were significantly increased(P <0.05);compared with the model group,the levels of VCAM-1 in each medication group were significantly decreased(P <0.05);compared with the dose group,the levels of VCAM-1 in the middle-dose and high-dose groups of Chinese medicine decreased(P <0.05).2.IL-1β:Compared with the control group,the levels of IL-1β in the model group and each medication group were significantly increased(P <0.05);compared with the model group,the levels of IL-1β in each medication group were significantly reduced(P <0.05);compared with the dose group,the levels of IL-1β in the middle-dose and high-dose groups of Chinese medicine decreased(P <0.05).3.Observation by light microscope showed that the nucleus of the model group was swollen and rounded,the myocardial interstitial edema,and the tissue gap widened.The nucleus of rats in the Chinese medicine group showed no obvious swelling,interstitial edema was alleviated,and myocardial cells were arranged densely and neatly.Conclusion: 1.Myocardial ischemia-reperfusion injury significantly increased the levels of VCAM-1 and IL-1β in the serum of rats;2.Shuangshen Tongmai Granules can reduce the levels of VCAM-1 and IL-1β in the serum of rats,and it is positively related to the drug dose;3.Shuangshen Tongmai Granules can protect the structure of myocardial tissue and cells.The possible mechanism is due to the reduction of VCAM-1 and IL-1β levels in rats. |
