ObjectiveTo investigate the role of Wenyang Tongmai Fang in rat myocardialischemia-reperfusion injury in vivo and explore its signal transduction pathway.Methodseighty-four male SD rats were randomly divided into shamegroup,myocardial ischemia-reperfusion injury group(IR group),WenyangTongmai Fang group and Wenyang Tongmai Fang plus blocking agent(LY294002,block PI3K-Akt signal transduction pathway)group. Shamegroup and IR group by gavage of saline for two weeks. Wenyang TongmaiFang group and Wenyang Tongmai Fang plus blocking agent group by gavageof Wenyang Tongmai Fang for two weeks. All rats,except for those in shamegroup, received occlusion of left anterior descending artery (LAD) for30minutes followed by120minutes of reperfusion,LY294002(0.3mg/kg)intravenous administration before reperfusion.Serum CK-MB were detectedafter experiment.Myocardial infarction size was detected by nitroblue tetrazolium staining method. Apoptotic cells were quantified by TUNEL method, andthe expression of Akt and P-Akt were determined by Western blotting.ResultsCK-MB,Apoptosis index and the expression of P-Akt of Shame groupwere significantly lower than those in IR group, Wenyang Tongmai Fang groupand Wenyang Tongmai Fang plus blocking agent group.CK-MB of Wenyang Tongmai Fang group was significantly lower than those in IR group(2587.29±241.57vs5216.68±387.76,P﹤0.05) and Wenyang Tongmai Fangplus blocking agent group(2587.29±241.57vs4016.72±219.34,P﹤0.05).Myocardial infarction size of Wenyang Tongmai Fang group was significantlylower than those in IR group (0.66±0.02vs0.83±0.04,P﹤0.05)and WenyangTongmai Fang plus blocking agent group(0.66±0.02vs0.76±0.03, P﹤0.05).Apoptosis index of Wenyang Tongmai Fang group was significantly lower thanthose in IR group(13.24±3.69vs29.76±5.37,P﹤0.05) and Wenyang TongmaiFang plus blocking agent group(13.24±3.69vs22.89±4.83,P﹤0.05). Theexpression of P-Akt in Wenyang Tongmai Fang group was significantlyincreased than that in IR group(0.57±0.08vs0.22±0.03,P﹤0.05) and WenyangTongmai Fang plus blocking agent group(0.57±0.08vs0.31±0.05,P﹤0.05).ConclusionWenyang Tongmai Fang may protect rat hearts from in vivo ischemia-reperfusion injury, attenuate myocardial infarction size and apoptosis, whichmaybe mediated by the activation of PI3K-Akt signal transduction pathway.
