Purpose : This experiment was to observe the effect of Shuangshen Huoxue granule(shhxg),which has the functions of Supplementing Qi and nourishing Yin,promoting blood circulation and dredging collaterals,on acute myocardial ischemia(AMI)rats,and to detect the effects of myocardial infarction area and the expression of autophagy related factors LC3,p62 and beclin-1,To provide a new theoretical basis for the treatment of acute myocardial ischemia of coronary heart disease with traditional Chinese medicine.Material and method:Ten of sixty rats were randomly selected as blank group,and the remaining fifty rats were ligated proximal left anterior descending coronary artery to establish acute myocardial ischemia model.The successful rats were randomly divided into model group,high,medium and low dose groups of traditional Chinese medicine(Shuangshen Huoxue Granules)and Western Medicine(trimetazidine tablets)groups.The blank group and model group were given normal saline by gavage.Two weeks later,the myocardial tissues of rats in each group were taken.The m RNA expressions of beclin-1,LC3 and p62 were detected by RT-PCR.The protein expressions of beclin-1,LC3 ? / LC3 ? and p62 were detected by Western blot.The infarct size was determined by TTC staining.Results:1.Compared with the blank group,the expression levels of beclin-1 gene and protein in the model group were increased(P < 0.05);LC3 gene expression increased(P < 0.05);The expression of LC3? / LC3?protein was also increased(P < 0.05);The expression of p62 gene and protein decreased(P < 0.05);The size of myocardial infarction increased(P< 0.05).2.Compared with the model group,the expression levels of beclin-1 gene and protein in the high,medium and low dose groups of Shuangshen Huoxue granule and the western medicine group were decreased(P < 0.05,P < 0.01);LC3 gene expression decreased(P< 0.05,P < 0.01);The expression of LC3? / LC3? protein also decreased(P < 0.05,P< 0.01);The expression of p62 gene and protein increased(P < 0.05,P < 0.01);The infarct size decreased(P < 0.05).3.Compared with the western medicine group,the expression of beclin-1,LC3 and p62 m RNA and the protein expression of beclin-1,LC3? / LC3?and p62,as well as the percentage of myocardial infarction area in the high-dose Chinese medicine group were not statistically different(P > 0.05).Conclusion:1.After acute myocardial ischemia in rats,the ischemic cardiomyocytes will have excessive autophagy,and the myocardial tissue will appear necrotic area.2.Shuangshen Huoxue granule and trimetazidine tablets can regulate the expression of beclin-1,LC3,LC3 ? / LC3 ? and p62,which can inhibit the autophagy activity of ischemic cardiomyocytes in rats with acute myocardial ischemia,and the inhibitory effect of high dose of traditional Chinese medicine is close to that of Western medicine.3.Shuangshen Huoxue Granules and trimetazidine tablets can reduce the size of myocardial infarction and the degree of myocardial injury,and the reduction degree of high dose of traditional Chinese medicine is similar to that of Western medicine.4.Shuangshen Huoxue Granules and trimetazidine tablets can improve the blood supply of myocardium and reduce the area of myocardial injury by regulating the autophagy activity of cells during acute myocardial ischemia.
