| Purpose:In this study,the effects of Shuangshen Tongmai Granule on myocardial ischemia-reperfusion injury and myocardial ischemia in rats were observed by observing the pathological structure of myocardial cells and detecting the levels of inflammatory factors IL-1 and TNF-?,providing ideas for further clinical applications.Material and method:1.Grouping and administration of experimental animals:SD rats were randomly divided into 5groups: normal saline control group,model group,positive control group(Western medicine group),high-dose Shuangshen Tongmai granule group(high-dose Chinese medicine group),low-dose double-shenzhen Tongmai Particle group(low dose Chinese medicine group).The model group and the blank control group were given normal saline 3ml/d,and the western medicine group was given 4.63 mg/(kg.d).The solution was mixed with normal saline to form a 3 ml solution.The high and low dose Chinese medicine groups were given double ginseng.Tongmai Granules 13,45 mg /(kg.d),37.5mg /(kg.d),respectively,were mixed with physiological saline to prepare 3ml of saline solution for intragastric administration,and were continuously administered for 4 weeks and then molding.2.Experimental animal modeling:The model group,the high-and low-dose Chinese medicine group,and the western medicine group were subjected to ligation at the 2 mm left anterior descending of the left coronary artery for 30 min,and then the ligature was opened to establish a myocardial ischemia-reperfusion model.The blank control group only opened the chest and was not ligated,and the model was established.Close the chest after success3.Specimen collection and index detection:After sacrifice,the myocardial tissue of the ligation site was taken for HE staining to observe the pathological changes,and the blood of the abdominal aorta was collected.The inflammatory factors interleukin IL-1 and TNF-?were determined according to the instructions of the relevant test kit.The content and the preventive effect and mechanism of Shuangshen Tongmai Granule on myocardial ischemia-reperfusion injury were observed and statistically analyzed.Results:1.Histopathological observation of HE staining: no significant degeneration of myocardial tissue in the control group,regular arrangement of myocardial fibers,and lighter interstitial edema;partial myocardial focal necrosis and myocardial fibrosis in the model group;high-dose Chinese medicine group and low-dose Chinese medicine group And the western medicine group showed that the myocardial fibers were arranged more regularly than the model group,the fractures were less,the interstitial edema was lighter,and the myocardial interstitial was slightly hemorrhagic.2.Serum IL-1 and TNF-? levels: IL-1 and TNF-? were slightly expressed in the blank control group;IL-1 and TNF-? were expressed in the model group;compared with the model group,the Western medicine group and the Chinese medicine group IL-1,The expression of TNF-? was decreased to some extent,and the expression level was the lowest in the high-dose Chinese medicine group.It indicated that Shuangshen Tongmai Granule can reduce the release of IL-1 and TNF-?.Conclusion:1.Shuangshen Tongmai Granule can improve the structure of myocardial cells after ischemia-reperfusion2.Shuangshen Tongmai Granules protect the myocardium by inhibiting the expression of inflammatory factors IL-1 and TNF-?. |
