ObjectiveTo observe the expression of mitochondrial autophagy related proteins NF-?B,p62 and parkin in colon tissue of ulcerative colitis model rats,and observe the activities of mitochondrial autophagy in colon tissue directly by transmission electron microscope,and to explore the effect and possible mechanism of regulating mitochondrial autophagy by Potentilla discolor.Method1.Pharmacodynamic evaluation of Potentilla discolor on UC model ratsTNBS/ethanol modified composite method was used to establish the model,and the ethanol extract of Potentilla discolor was given continuously for 14 days.Draw the trend chart of DAI score of experimental rats for interpretation,observe and compare the pathological changes of colon tissue in each group of experimental rats,and comprehensively analyze the effect of Potentilla discolor on intestinal mucosa repair and treatment of UC model rats.2.Study on the mechanism of mitochondrial autophagy in rats with ulcerative colitisThe expression of NF-?B,p62 and parkin was detected by Western blot and qRT-PCR.The autophagy of mitochondria in the colon was observed by transmission electron microscopy.The role of mitochondrial autophagy in the occurrence and development of ulcerative colitis was preliminarily clarified,and the possible mechanism of repair of colonic mucosa damage by regulating mitochondrial autophagy was discussed.Results1.Through the comparative analysis of DAI score trend chart and colonic pathological morphology of rats in each group,we found that Potentilla discolor group and sulfasalazine group had better effect in improving the inflammation of UC model rats and repairing the damaged intestinal mucosa of UC model rat.2.The results of qRT-PCR showed that there was significant difference in Parkin,NF-?B and p62 expression between the model group and the blank group(P<0.05).The expression'of Parkin,NF-?B and p62 in the model group was statistically significant(P<0.05)after 14 days of continuous gavage.3.The results of transmission electron microscopy showed that the mitochondria in the group of Potentilla discolor were swollen and the fusion of mitochondria and lysosomes was obvious,and more lysosomes could be found around the mitochondria.It indicated that there was obvious mitochondrial autophagy in Potentilla discolor group.Conclusion:1.The effect of Potentilla discolor on ulcerative colitis in TNBS/ethanol modified compound model rats was better than that of sulfasalazine.2.Potentilla discolor can play a therapeutic role in UC by regulating the expression of mitochondrial autophagy related proteins in colon mucosa of UC model rats.
