| Objective:To observe the effect of electroacupuncture at the acupoints of Shao yang Meridian on 5-Hydroxy Tryptamine(5-HT),Substance P(SP),and Calcitoni n Gene Related Peptide(CGRP)levels and 50% Paw Withdrawal Threshold(50%PWT)in a rat model of Nitroglycerin(NTG)-induced migraine(ME),and to compare the therapeutic efficacy of electroacupuncture at the acupoints of Sha oyang Meridian and Non-Meridian/Acupoint in the treatment of NTG-ME rats,t o explore the possible mechanism of electroacupuncture at the acupoints of Sha oyang Meridian for migraine.Methods: Forty healthy Sprague-Dawley rats were randomly divided into four groups: blank group,model group,electroacupuncture at the acupoints of Shaoy ang Meridian treatment group(the acupoints of Shaoyang Meridian group)and electroacupuncture at the Non-Meridian/Acupoint treatment group(Non-Meridian/Acupoint group).with 10 rats in each group.Model group,the acupoints of Sha oyang Meridian group and Non-Meridian/Acupoint group according to 10mg/kg hip subcutaneously the NTG building,hip subcutaneous injection amount of sal ine blank group,30 min after building,the acupoints of Shaoyang Meridian gr oup and Non-Meridian/Acupoint group 30 min line corresponding cupping treat ment respectively,measuring between groups of rats at different stages of 50% PWT,using ELISA in serum of rats was detected in 5-HT,SP and CGRP lev els.Results: 1.After modeling,NTG-ME rats showed redness in both ears,the forelimbs fre quently scratched their heads,increased the number of cage climbing,and Incr eased performance of vertical hair,tail biting and other activities.The behavior al scores of NTG-ME rats increased and the 50%PWT decreased(P<0.01),indi cating that the modeling was successful;2.To compare the 50%PWT in each group:After treatment,compared with afte r modeling,the 50% PWT of the acupoints of Shaoyang Meridian group and N on-Meridian/Acupoint group were significantly increased(P<0.01).Compared with the model group,there was no significant difference in 50%PWT in the Non-Meridian/Acupoint group(P>0.05),and the 50%PWT in the Shaoyang Meridian group was significantly higher(P<0.01).Compared with the Non-Meridian/Acup oint group,the 50%PWT of the Shaoyang Meridian group was significantly hig her(P<0.01).3.To compare the contents of 5-HT,SP and CGRP in each group: Compared with the model group,the serum level of 5-HT in the acupoints of Shaoyang Meridian group was significantly increased(P<0.01),and the contents of SP an d CGRP were significantly decreased(P<0.01).There was no statistical signific ance in the Non-Meridian/Acupoint group(P>0.05).Compared with the Non-Mer idian/Acupoint group,the content of 5-HT in Shaoyang Meridian group decreas ed significantly(P<0.01),and the content of SP and CGRP increased significan tly(P<0.01).Conclusion: 1.Electroacupuncture at the acupoints of Shaoyang Meridian can alleviate the p ain degree of NTG-ME rats,significantly increase the content of 5-HT in seru m,effectively down-regulate the expression of SP and CGRP in rat serum,whi ch may be one of the mechanisms of action in the treatment of ME rats;2.The therapeutic effect of electroacupuncture at the acupoints of Shaoyang M eridian on NTG-ME rats is better than that of electroacupuncture at the Non-M eridian/Acupoint.which provides some experimental basis for the clinical select ion of electroacupuncture at the acupoints of Shaoyang Meridian for treating M E and the specificity study of acupuncture effect. |
