Objective:The detection of Yifeijianpi Formula in high,medium and low three groups of the chronic obstructive pulmonary disease rat model in bronchial lung tissue Toll-like receptors(TLR4),nuclear factors-?B(NF-?B)and Mucin 5AC(MUC5AC),protein expression of Yifeijianpi Formula in each dose;Investigate the mechanism of Yifei Jianpi Formula in the treatment of chronic obstructive pulmonary airway mucus hypersecretion and whether Yifei Jianpi Formula can through the TLR4/NF-?B/MUC5 AC pathway to control COPD airway mucus hypersecretion.Methods:112 male SD rats were randomly divided into 7 groups: normal group,model group,Yifei Jianpi Formula high,middle and low dose group,ambroxol group and dexamethasone group,with 16 rats in each group.Except for 16 rats in the blank group,96 rats in the other groups were treated with smoked,intratracheal instillation of lipopolysaccharide and senna leaf to establish the rat model of chronic obstructive pulmonary,spleen and spleen deficiency.The pathological changes of lung and bronchus in each group were observed by HE staining.The bronchial goblet cells in each group were observed by AB-PAS staining.The protein expression levels of TLR4,NF-?B and MUC5 AC in bronchopulmonary tissue of rats were detected by Western blot.fq RT The expression of TLR4 m RNA,NF-?B m RNA and MUC5 AC m RNA in bronchopulmonary tissue of rats were detected by PCR.The expression of TLR4,NF-? B and MUC5 AC in bronchopulmonary tissue of rats was detected by immunohistochemistry.Results: 1.Histomorphological observation: HE staining display that the rats of COPD model group,s Bronchial and lung tissue HE staining comply with COPD lung tissue pathology.In the Yifei Jianpi treatment group,the ambroxol treatment group and the dexamethasone treatment group had different degrees of lung tissue injury compared with the normal control group,but the degree of injury was significantly reduced compared with the COPD group.the most obvious reduction was in the high dose roup of Yifu Jianpi Formula and the dexamethasone group.AB-PAS staining showed that there were a large number of goblet cell hyperplasias in the bronchus of the model group;a small amount of goblet cell hyperplasia was seen in the dexamethasone group ?ambroxol group and each dose group of Yifei Jianpi Formula.the number of goblet cells was significantly reduced when it was compared with the control group.2.The expression of TLR4,NF-?B and MUC5 AC in bronchopulmonary tissue of rats in each group(1)Western blot method: compared with the normal group,the levels of TLR4,NF-?B,MUC5 AC protein in the model group increased significantly(P<0.01);compared with the model group,each dose group of Yifei Jianpi Formula,ambroxol group and dexamethasone group was decreased(P<0.05);compared the expression of TLR4,NF-?B,MUC5 AC in each dose group of Yifei Jianpi Formula,high dose group was the lowest;there was no obvious difference in the Yifei Jianpi high dose group compared with the ambroxol group and dexamethasone group(P>0.05).(2)Reverse transcriotion-polymerase chain reaction: compared with the normal group,the levels of TLR4 m RNA,NF-?B m RNA and MUC5 ACm RNA in the model group rats was increased(P<0.01);compared with the model group,the levels of TLR4,NF-? B,MUC5 AC in each dose group of Yifei Jianpi Formula,ambroxol group and dexamethasone group was decreased(P<0.05);compared the expression of TLR4,NF-?B,MUC5 AC protein content in each dose group of Yifei Jianpi Formula,high dose group was the lowest;there was no obvious difference in the expression of NF-?B m RNA and MUC5 AC m RNA in the Yifei Jianpi high dose group compared with the ambroxol group and dexamethasone group(P>0.05).(3)Immunohistochemistry: compared with the normal group,the levels of TLR4,NF-?B and MUC5 AC in the model group rats was increased(P<0.01);compared with the model group,the levels of TLR4,NF-?B and MUC5 AC in the high?middle dose group of Yifei Jianpi Formula,ambroxol and dexamethasone decreased(P <0.01,P <0.05);compared with the model group,the level of TLR4 decreased in the low ose group(P<0.01);compared of each dose group of Yifei Jianpi Formula,high dose group was the lowest;the expression of TLR4 and MUC5 AC in the high dose group of Yifei Jianpifang was obviously higher than that in the ambroxol and dexamethasone groups(P<0.01,P<0.05;but no obvious difference was found between between the Yifei Jianpi high dose group and the ambroxol group(P>0.05),but Yifei Jianpi high dose group was obviously higher than that of the dexamethasone group(P<0.01).Conclusion: Yifeijianpi Formula can improve the pulmonary function in COPD model rats and reduce pathological damage,decrease the expression of TLR4,inhibit the activation of NF-B pathway,regulate and control the transcription and expression of adhesion protein of MUC5 AC,through reducing the inhibition of airway inflammation and airway mucus hypersecretion to achieve the purpose of preventing COPD.
