Objective:Literatures study to clarify the efficacy of sanziyangqin decoction for asthma.Establish the best way for sanziyangqin decoction in water.The study of asthmatic rats mucus hypersecretion mechanism controlled by Sanzi Yangqin Decoction based on CysLTs pathway.Method:Research the RCTs that published in biomedical journals use sanziyangqin decoction to cure the asthma.Rev Man5.3 was used to process data.Best water extraction is choosed by three-dimensional effect surface.Sixty SD rats were randomly divided into blank group,model group,and four treatment groups.Rats will be injected into the abdomen with OVA and atomized by ultrasound attack with 2ml OVA NS once a day(Half an hour a day for 14 days).Rats in each grounp receive gavage of 1ml that containing specific drug.After the sensitization,tail vein pumping for each rats at 8am and 8pm.Rats were sacrificed at 28~thh day.Serum and tissue samples were collected for detecting the content of muc5ac?CysLTsR1?P38MAPK?P-P38MAPK protein and so on.Result:1.The difference between Z=5.36(P<0.0001)was statistically significant?2.The best extraction process of the sanziyangqin decoction is 12 times solvent multiple,extraction 2 times,each time 1.2h.3.The rats in the blank group were strong and responsive,with smooth and smooth fur,good drinking water and significant weight gain.Simply use OVA sensitization and untreated rats 7 days start activity decreases,the 14th day,shortness of breath,slow,curled up,outlining the fur sparse lacklustre,feeding water into a downward trend,sensitization after weight gain in rats was very slow.The other four rats were treated with drug therapy group,and the mental state was better and the activity frequency was increased compared with the model group,and the intake of water was close to the blank group,and the weight gain was faster.4.The normal group alveolar tissue formation rules,the lumen wall is smooth,no inflammatory cells infiltrate and ooze.In the model group,a large number of inflammatory cells were infiltrated,the alveolar wall was irregular,accompanied by epithelial cell cup hyperplasia,the local basement membrane thickened,and the necrosis tissue was seen in the alveolar cavity.The treatment group still had local inflammatory infiltration,pulmonary alveolar and endobronchial glandular hyperplasia was not active,and there was still a small amount of edema and exudation,and there was a little necrotic obstruction in the small airway.5.?TNF-?:Compared with the control group,alveolar lavage fluid at each time point in the model group(BALF)in TNF-?levels increased were significantly different(P<0.05).Compared with the model group,montelukast group and each time point sanziyangqin decoction high,medium and low dose group were lower alveolar lavage fluid(BALF)in TNF-?content were significantly different(P<0.05)?IgE:Compared with the control group,alveolar lavage fluid at each time point in the model group(BALF)in IgE levels increased were significantly different(P<0.05).Compared with the model group,montelukast group and each time point sanziyangqin decoction high,medium and low dose group were lower alveolar lavagefluid(BALF)in IgE content were significantly different(P<0.05)?LTC4:Compared with the control group,alveolar lavage fluid at each time point in the model group(BALF)in LTC4 levels increased were significantly different(P<0.05).Compared with the model group,montelukast group and each time point sanziyangqin decoction high,medium and low dose group were lower alveolar lavage fluid(BALF)in LTC4 content were significantly different(P<0.05).Compared with the montelukast grounp,sanziyangqin decoction high,medium in LTC4 content were not significantly different P>0.05,low dose group P<0.05.6.The expression of MUC5AC?CysLTsR1?P38MAPK?PP38MAPK compared with the control group,the expression of MUC5AC?CysLTsR1?P38MAPK?PP38MAPK in lung tissue of rats in other group increased were significantly different(P<0.01).Compared with the model group,the cured grounp the expression of MUC5AC?CysLTsR1?P38MAPK?PP38MAPK in lung tissue of rats in other group increased were significantly different(P<0.05).7.the expression of Muc5ac mRNA Compared with the control group,the expression of MUC5ACmRNA in lung tissue of rats in other group increased were significantly different(P<0.05).Compared with the model group,the cured grounp the expression of MUC5ACmRNA in lung tissue ofrats in other group increased were significantly different(P<0.05).In each cured grounp,sanziyangqin high does grounp perform better.Conclusions:1.the effect of sanziyangqing decoction and western medecine is bttter than only wstern medecine.2.The best extraction process of the sanziyangqin decoction is 12 times solvent multiple,extraction 2 times,each time 1.2h.3.To make a asthma mucus hypersecretion rat model through OVA intraperitoneal injection and ultrasonic atomization is successful.4.Sanziyangqin decoction can effectively inhibit the expression of MUC5AC in lung tissue,and TNF-??Ig-E?LTC4 in serum.5.Sanziyangqin decoction can effectively inhibit the expression of CysLTSR1 and inhibit the phosphorylation of P38MAPK,that can regulate MUC5AC protein in transcriptional levels.
