Objective: In order to explore the mechanism of Bufei Decoction in the treatment of chronic obstructive pulmonary disease emphysema from the perspective of immune mechanism,we established the animal model of chronic obstructive pulmonary disease emphysema and intervened with Bufei Decoction to detect the expression of Foxp3,IFN-? and IL-4.Methods:42 SPF SD rats(all male)were randomly divided into blank group,model group,spleen Aminopeptide group and Bufei Decoction group.There were 12 in model group and 10 in other groups.Except for the blank group,the other groups were given intratracheal injection of lipopolysaccharide combined with cigarette passive smoke to make COPD emphysema animal model.At the same time,the blank group was injected with the same amount of normal saline as lipopolysaccharide into the trachea without passive fumigation.After the model was established successfully,the blank group and the model group were given normal saline,Bufei Decoction group and spleen Aminopeptide group respectively.After treatment,the expression of Foxp3 was detected by RT-q PCR and the levels of IFN-? and IL-4 in serum and BALF were detected by ELISA.Results:1.Compared with the model group,the expression of Foxp3 in lung tissue of Bufei Decoction group increased(P < 0.01).There was no significant difference in the content of Foxp3 between Bufei Decoction group and spleen Aminopeptide group(P > 0.05).2.Compared with the model group,the content of IFN-? and IL-4 in the serum and BALF of Bufei Decoction group was significantly different(P < 0.01);compared with the spleen Aminopeptide group,the content of IFN-? and IL-4 in the serum and BALF of Bufei Decoction group had no significant difference(P > 0.05).Conclusion:Bufei decoction can increase Foxp3 expression,increase IFN-? and decrease IL-4 content,regulate T cell-mediated acquired immunity,improve immune ability of rats,improve airway damage,alleviate alveolar expansion and pathological fusion,so as to achieve the purpose of treating COPD emphysema.
