Effects Of Bufei Decoction On Pulmonary Vascular Endothelium In Rats With Chronic Obstructive Pulmonary Disease

Objective: Objective to establish a rat model of chronic obstructive pulmonary disease(COPD)and observe the changes of pulmonary vascular endothelial structure and function after intervention with Bufei decoction,so as to explore the mechanism of Bufei Decoction in treating COPD rats.Methods: A total of 32 clean grade SD rats were randomly divided into blank group,model group and traditional Chinese medicine group.There were 10 rats in the model group and 10 rats in the other two groups.The COPD model was reproduced by smoking combined with lipopolysaccharide.After successful modeling,the original formula of Mianjian granule Bufei decoction was given by gavage on the 29 th day,and the related specimens were collected after 15 days of continuous administration.During the whole experiment,the general state and behavior of rats were observed dynamically.One day after the last gavage,blood and lung tissue samples were collected from abdominal aorta of rats.The contents of ET-1,NO,tPA,PAI and vWF in serum were detected by ellsa;the pathological changes of pulmonary arterioles were observed by HE staining,and the structure and morphology of vascular endothelial cells were observed by transmission electron microscope.Results:(1)General situation:compared with the model group,the Bufei Decoction group has better mental state,less irritability and excitement,slower respiratory rate and better appetite.(2)Pathological changes of rat pulmonary vessels: in the model group,the intima and vascular smooth muscle of pulmonary arterioles were obviously proliferated,neutrophil infiltration could be seen in the periphery of vascular wall;the arrangement of vascular endothelial cells was disordered and deformed,and exfoliated vascular endothelial cells could be seen in the lumen.The above pathological changes in Bufei Decoction group were less than those in model group,but still more serious than those in blank group.(3)Changes of serum ET-1,NO,tPA,PAI and vWF: 1)compared with the blank group,the levels of no and tPA in the model group were decreased(P < 0.05),while the levels of ET-1,Pai and vWF in the model group were increased(P < 0.05)The expression of PAI increased(P < 0.05),while the expression of ET-1,PAI and vWF decreased(P <0.05),indicating that Bufei decoction can improve the vascular endothelial function of COPD by regulating the imbalance of ET-1 / no and TPA / PAI,and reducing the content of vWF in serum;2)the results of electron microscopy showed that the gap between pulmonary vascular endothelial cells increased,the cell membrane collapsed,the nucleus fused,the mitochondria damaged obviously,and obvious edema or empty space could be seen Vesicular degeneration and disappearance of internal structure ridge.In the Bufei Decoction group,the degree of structural damage of pulmonary vascular endothelial cells was significantly reduced,and the mitochondria were significantly repaired.Conclusion:1.Bufei decoction can improve the structure and function of pulmonary vascular endothelium in COPD rats.2.Bufei decoction can improve the vascular endothelial function of COPD rats by regulating the imbalance of ET-1/NO and TPA /PAI and reducing the content of serum VWF.
