| Objective:To investigate the effects of Moxibustion at Guanyuan and Zusanli on the expression of inflammatory cytokines in the colon of rats with ulcerative colitis(UC)induced by dextran sodium sulfate(DSS),and to explore its related mechanisms.Methods:50 SD rats were fed for 3 days.According to the body weight,they were divided into 5groups,10 in each group: normal group,model group,western medicine group,moxibustion group,moxibustion plus western medicine group,caged,Except the normal group,the other 4groups of rats were free to drink the prepared 4% DSS solution for 7 days,while measuring and recording the color,weight and stool of the rat hair every day.The model is completed,treated,and fed normally.In the normal control group,10 mL/(kg.d)of normal saline was administered once a day.The model control group,normal saline 10 mL /(kg.d),once a day.Western medicine group,mesalazine aqueous solution 10 mL /(Kg.d),once a day gavage.In the moxibustion treatment group,the moxibustion group was treated with moxibustion after30 minutes of gavage,and the points were taken: Guanyuan and Zusanli(double).Ai Wei is made of refined cashmere and weighs about 90mg/piece.Each time,each moxibustion is 1strong,1 time/d.The moxibustion western medicine group was treated with mesalazine aqueous solution 10mL/(Kg.d)once a day;after 30 minutes of gavage,moxibustion was performed,and the moxibustion group was operated.At the same time,the color and stool of the rat hair were observed,and the body weight was measured and recorded.At the end of the experiment,the serum levels of tumor necrosis factor-?(TNF-?),IL-8,IL-13 and IL-10 were observed by double antibody sandwich ELISA.Reverse transcription-polymerase chain reaction(RT-PCR)was used.The expressions of IL-8 mRNA and IL-10 mRNA in intestinal mucosa were detected,and the effect of moxibustion on inflammatory cytokines in colon of UC rats was investigated.RESULTS:1.Compared with the control group,the weight of the rats after modeling was significantly reduced,there were blood in the stool,colonic mucosal edema,hemorrhage;elevated IL-8mRNA levels in the colonic mucosa and decreased IL-10 mRNA levels,all of which showed significant differences(P<0.05)was statistically significant;serum levels of TNF-?,IL-8,IL-13 were increased(P<0.05),and IL-10 levels were decreased(P<0.05).Compared with the control group,the Th1 cytokine was enhanced in the colonic mucosa of the model group,and the Th2 cytokine was inhibited.2.Compared with the model group,the weight of the rats in the treatment group increased significantly,the degree of blood in the stool was significantly improved,and the degree of colonic mucosal edema and hemorrhage was significantly reduced.3.Compared with the model group,the serum levels of IL-10 in the western medicine group,the moxibustion group and the moxibustion plus western medicine group were increased(P<0.05),and the levels of IL-13,TNF-? and IL-8 were down-regulated.(P<0.05),IL-8mRNA levels in intestinal mucosa decreased(P<0.05)and IL-10 mRNA levels increased(P<0.05),indicating that the immune imbalance on the colonic mucosa of rats was restored,and the effect of Th2 immune cells was enhanced.Immune cell action is inhibited.Conclusion:1.Moxibustion UC rats Guanyuan and Zusanli can reduce or relieve the mucus pus and bloody stool in UC rats.2.Moxibustion UC rats Guanyuan and Zusanli can promote the formation of anti-inflammatory factors in the colonic mucosa of UC rats,and inhibit the formation of pro-inflammatory factors,thereby alleviating intestinal mucosal damage. |
