Based On The TLR4/MyD88/NF-κB Signaling Pathway, The Effects Of Kuijiekang Pills On Th1/Th2 Immune Balance In UC Rats Were Investigated

Purpose: By detecting the protein content of TLR4,MyD88,NF-κB in the colonic mucosa and contents of TNF-,Il-8,Il-13 and Il-10 in serum in ulcerative colitis rats.To investigate whether Kuijiekangwan interfere with TLR4/Myd88/NF-κB signaling pathway to affect Th1/Th2 immune balance,thus treating ulcerative colitis.To explore the mechanism of action of traditional Chinese medicine Kuijiekangwan,and provide experimental basis for clinical treatment of ulcerative colitis.Material and method: 50 SD rats were randomly divided into 5 groups: blank group,model group,standard treatment group,Chinese medicine treatment group,and traditional Chinese medicine compound standard drug treatment group,with 10 rats in each group.The blank group was excluded,and other groups were induced by DSS to induce a rat model of ulcerative colitis.Rats in the blank group and the model group after modeling were intragastrically administered with physiological saline 10 mL/(kg.d)once a day.After standard model treatment group,Chinese medicine treatment group and traditional Chinese medicine compound standard treatment group were molded,respectively,with mesalazine suspension and(or)Kuijiekangwan suspension 10 mL /(Kg.d),once a day.All groups were continuously administered for 14 days.The weight and stool of the rats were recorded daily during the treatment.After the end of the treatment,all rats were anesthetized and laparotomy,blood was taken from the abdominal aorta,and then the degree of colonic mucosal lesions in each group was observed.The levels of serum Tumor Necrosis Factor-α(TNF-α),IL-8,IL-13 and IL-10 were detected by ELISA.The intestinal mucosal IL-8 was detected by Real time-Polymerase Chain Reaction(RT-PCR).The expression of mRNA and IL-10 mRNA was detected by Western Blot.The content and expression of TLR/MyD88/NF-κB protein in intestinal mucosa of UC rats were determined.Results: Compared with the rats in the blank group,the body weight of the rats after modeling was significantly reduced(P<0.05),the degree of blood in the stool increased,the edema and hemorrhage of the colonic mucosa appeared,and the DAI score increased.Thecontent of key protein in TLR/MyD88/NF-kB in colonic mucosa was significantly increased,the level of IL-8 mRNA in intestinal mucosa was increased,and the level of IL-10 mRNA was decreased.The results were statistically significant(P<0.05).The levels of TNF-α,IL-8and IL-13 in serum increased(P<0.05),and the levels of IL-10 decreased(P<0.05).The TLR/MyD88/NF-κB signaling pathway was activated in the colonic mucosa of the model group compared with the blank rats.The Th1 immune axis was enhanced and the Th2 immune axis was inhibited.After drug treatment,the weight of the rats in the treatment group increased significantly,the degree of blood in the stool was reduced,the degree of colonic mucosal edema and hemorrhage was also significantly improved,the DAI score was reduced,and the disease was improved.Compared with the model group,the levels of IL-10 in the serum of the standard treatment group,the Chinese medicine group,and the traditional Chinese medicine compound standard treatment group were increased(P<0.05),and the levels of IL-13,TNF-α,and IL-8 were down-regulated(P < 0.05).IL-8 mRNA levels in intestinal mucosa decreased(P<0.05)and IL-10 mRNA levels increased(P<0.05).The key protein of TLR/MyD88/NF-κB signaling pathway was detected by WB method(P<0.05).This indicates that the signaling pathway is inhibited,the Th2 immune axis is enhanced,and the Th1 immune axis is inhibited(P<0.05).In the comparison of the efficacy of these three groups,the traditional Chinese medicine compound standard treatment group was significantly better than the Chinese medicine group and the standard treatment group(P<0.05).Conclusion: In the colonic mucosa of rats with ulcerative colitis,the TLR4/Myd88/NF-κB signaling pathway is activated,and the Th1/Th2 immune balance is broken,and the direction of chemotaxis is progressing.At this time,the pathway may directly affect the pathological process of ulcerative colitis.The Chinese medicine Kuijiekangwan may regulate this signaling pathway,affect the Th1/Th2 immune balance axis,and play a therapeutic role in ulcerative colitis.At the same time,it was found that Kuijiekang Pill combined with mesalazine has the best therapeutic effect,and the combination may enhance the regulation of the signaling pathway.
