Purpose: To investigate the effect of Sangpi Zhike Granule on lung histopathology and expression of PI3K/AKT and gamma-GCS in guinea pigs with allergic cough.Material and method:Sixty healthy guinea pigs aged 8 weeks of SPF grade were randomly divided into normal control group,model control group,western medicine control group(montelukast sodium),low-dose group of mulberry peel cough granule,medium-dose group of mulberry peel cough granule and high-dose group of mulberry peel cough granule,with 10 guinea pigs in each group.Except for the normal control group,allergic cough model was established by ovalbumin sensitization in other groups.After gastric lavage,10% chloral hydrate(3.3ml/Kg)was injected intraperitoneally into each group of guinea pigs according to their body weight.The splint was fixed and dissected.Serum and lung tissues were collected.Immunohistochemical staining of P-AKT and gamma-GCS in lung tissue was used to observe the frontal cortex of guinea pigs in each group under light microscope,and images were collected.PI3 K and gamma-GCS were detected by RT-PCR.Results:1.Compared with the symptoms of guinea pigs,the model guinea pigs had cough and other symptoms.After treatment,all groups of guinea pigs were improved,and there was no significant abnormality in the normal control group.2.The pathological comparison of the lungs of guinea pigs showed that the epithelial cells of the normal control group were relatively intact and the lung structure was normal;the epithelial cells of the model group were severely exfoliated and necrotic,which might be accompanied by severe inflammatory cell infiltration.The epithelial exfoliation,necrosis and inflammatory cell infiltration of the treated groups were lighter than those of the model control group.3.At the end of the experiment,there was no death in the normal group,the western medicine treatment group,the middle and high dosage groups of Sangpi Zhike Granule,while there was one death in the model control group and one death in the low dosage group.4.Compared with the normal group,the number of coughs in the model control group and each treatment group increased,and the latent period of cough shortened,with significant difference(P < 0.05);Compared with the model control group,the number of coughs in the western medicine control group and the mulberry cough relieving granule groups decreased significantly,and the latent period of cough prolonged,with significant difference(P < 0.05);compared with the western medicine control group,the cough granule low and medium dose groups coughed significantly(P < 0.05).There was no significant difference between the high dose group and the low dose group(P > 0.05).Compared with the high dose group,the low and middle dose groups had more cough and shorter cough latency(P < 0.05).5.Compared with the normal group,the P-AKT protein and gamma-GCS protein in the model control group and each treatment group were higher,with significant difference(P < 0.05);Compared with the model control group,the P-AKT protein and gamma-GCS protein in the western medicine control group and the Sangpi Zhike granule groups were lower,with significant difference(P < 0.05);Compared with the Western medicine control group,the P-AKT protein and gamma-GCS protein in the lower and middle dose groups were higher(P < 0.05).There was a significant difference(P < 0.05),but there was no significant difference(P > 0.05)between the high-dose group and the low-dose group.Compared with the high-dose group,P-AKT protein and gamma-GCS protein were higher in the low-dose group and the middle-dose group,and there were significant differences(P < 0.05).6.Compared with the normal group,the PI3 K and gamma-GCS m RNA in the model control group and each treatment group were higher,with significant difference(P < 0.05);compared with the model control group,the PI3 K and gamma-GCS m RNA in the western medicine control group and the mulberry antitussive granule dose groups were lower,with significant difference(P < 0.05);compared with the western medicine control group,the PI3 K and gamma-GCS m RNA in the lower and middle dose groups were higher,with significant difference(P < 0.05).There was no significant difference(P > 0.05)between the high-dose group and the low-dose group(P < 0.05).Compared with the high-dose group,PI3 K and gamma-GCS were higher in the low-dose group and the middle-dose group(P < 0.05).7.Compared with the normal group,the model control group and each treatment group had higher GSH and ROS and lower T-AOC,with significant difference(P < 0.05);compared with the model control group,the western medicine control group and the mulberry cough relieving granule dose groups had lower GSH and ROS and higher T-AOC(P < 0.05);compared with the western medicine control group,the cough granule group had higher GSH and ROS and lower T-AOC,with significant difference(P < 0.05);compared with the western medicine control group,the cough granule group had lower T-AOC and higher T-AOC.There was no significant difference(P > 0.05)between the two groups(P < 0.05).Compared with the high dose group,the low and middle dose groups had higher GSH and ROS and lower T-AOC(P < 0.05).Conclusion:1.Sangpi Zhike Granule can significantly improve the general state of allergic cough guinea pig model,alleviate cough symptoms,alleviate lung pathological changes,and play a role in the treatment of allergic cough.2.Sangpi Zhike Granule can regulate the expression of gamma-GCS by affecting PI3K/AKT signaling pathway,so as to significantly improve the oxidative and antioxidant imbalance caused by allergic cough,and thus play an antioxidant role.3.Sangpi Zhike Granule can significantly alleviate the infiltration of inflammatory cells and remove ROS to play an anti-inflammatory and antioxidant role,providing experimental basis for the clinical application of Sangpi Zhike Granule.
