Effect Of Mulberry Cough Granule On IL - 5 And EOS Levels In Guinea Pigs With Allergic Cough

Purpose:Using mulberry Zhike granules to treat irritability cough model of guinea pigs, by observing the guinea pig cough, lung tissue pathology, IL- 5 levels, cell count in BALF, explore the mulberry Zhike granules efficacy and mechanism of treatment of allergic cough, provide experimental basis for the further clinical application.Material and method:1、By intraperitoneal injection of allergic cough guinea pig model: the egg albumen sensitization and atomization inhalation of egg albumin arouse cough, the method of establishing juvenile guinea pig allergic cough model, through comparing the general situation of guinea pigs, detection of lung tissue pathology, counting capsaicin exciting cough and the cell counts in BALF, appraisal model of the guinea pig allergic cough. 2、Mulberry Zhike granules for the treatment of allergic cough guinea pigs effect and mechanism of research: on the basis of the success to establish animal model of allergic cough, set up the normal control group, model control group, the western medicine control group, mulberry Zhike particles in low dose group, mulberry Zhike particles dose group, the mulberry Zhike particles high-dose group and observation group of guinea pigs in general, the differences between the appearance and histopathological changes in lung tissue, leukocyte count and classification of EOS in BALF counting, IL- 5 levels in serum and BALF, explore the mechanism of action of mulberry skin allergic cough cough granules treatment.Results:1、Compared with normal group, model group of guinea pigs with poor general condition, cough obviously increase, significant difference(P < 0.05),Lung tissue pathology also have obvious change;white blood cell count and EOS in BALF percentage is high, the significant difference(P < 0.05),preparation success showed that allergic cough model of guinea pig. 2、Guinea pig general state observation:Normal group of guinea pigs in good condition,Model group of guinea pigs in bad condition,the condition in each treatment groups is improved. 3、Lung tissue pathology detection:Normal group of guinea pig lung tissue is without pathological changes; Model group presented the pathological changes; Lung lesions in each treatment group were compared with model group. 4、Cough count:Cough model group was obviously more than the normal group, the difference was statistically significant(P < 0.05); All medicine-consumption cough less than the model group, the difference has statistical significance(P < 0.05); Mulberry Zhike granules, middle and high dose group compared with the western medicine group, there was no statistically significant difference(P > 0.05), mulberry Zhike granuleslow dose group compared with it, the cough counts is more,there was the significant difference(P < 0.05). 5、Immunohistochemical and ELISA results of IL-5 showed:content of model group obviously higher than normal group, the difference was statistically significant(P < 0.05); Content of each treatment group compared with model group, the significant difference(P < 0.05); Mulberry Zhike granules, middle and high dose group compared with the western medicine group, there was no statistically significant difference(P > 0.05), mulberry Zhike granules low dose group compared with significant difference(P < 0.05). 6、White blood cell count and EOS in BALF percentage results show:Compared with normal group, model group significant difference(P < 0.05); Each treatment group compared with model group, significant difference(P < 0.05); Mulberry Zhike granules, middle and high dose group compared with the western medicine group, there was no statistically significant difference(P > 0.05), mulberry Zhike granules low dose group compared with significant difference(P < 0.05).Conclusion:1.With egg albumen sensitization and atomization inhalation arouse cough c be successfully established anaphylactic cough model of guinea pigs. 2.Mulberry Zhike granules could improve the symptoms of cough, relieve inflammation of lung tissue, mechanism may be related to regulating involv in immune inflammation cytokines IL- 5 related to the level of EOS.
