According to the knowledge of gynecology of TCM, pediatric coughing is determined by physiological characteristics of children. Due to the asthenia of spleen-stomach, children are easily affected by milk, cold and heat, causing the dysfunction of the spleen in transport. In this case, food essence cannot be absorbed very well and the obstruction of the lung-qi leads to cough and asthma.It will bring a lot of pains for children.Traditional Chinese medicines are effective for Pediatric coughing, which always act through various pathways.They are usually mild,with few side effects.They can treat both symptoms and causes of disease,with regulation on generalized condition.There are many famous classic formulas in ancient times,which can be prepared into Chinese patents by modern techniques.These patent are effective as ancient in clinic,and is convenient to use.Xiaoer Kechuan Granule stems from Professor Liu’s clinical experiences, which contains ten kinds of Chinese herbs like Honeysuckle, Lily Magnolia and the others. The granule is significantly effective in dispelling wind, clearing heat, relieving sore throat, ventilating lung, preventing cough and asthma, applied to avoid the failure of ventilating lung. Symptoms such as coughing, cough and asthma, sticky yellow phlegm, nasal obstruction, fever and aversion to wind, sweeting, throat-swelling, headache, red tongue, thin yellow tongue coating, floating and rapid pulse can be seen in clinic.Considering the function of improving coughing and characteristics of pediatric coughing, we studied its pharmacologieal effect by conventional animal models. Various functions were observed:antitussive, expectorant, decrease of the airway resistance and airway inflammation, in order to provide sufficient experimental data for the clinical using.Main research contents and results were as follows:1. Study on antitussive effects of Xiaoer Kechuan GranuleObjective:To observe the antitussive effects of Xiaoer Kechuan Granule. Methods: Antitussive effects were observed through cough test induced by SO2steaming and citric acid, after administration for successive there days.. Results:compared with control group, Xiaoer Kechuan Granule at the dosage of9g/kg,4.5g/kg,2.25g/kg could decrease the frequency of mice cough induced by ammonia. Xiaoer Kechuan Granule at the dosage of9g/kg,4.5g/kg,2.25g/kg could decrease the frequency of guinea pigs cough induced by citric acid(P<0.05or P<0.01). Conclusion:Xiaoer Kechuan Granule showed certain antitussive effects. 2. Study on expectorant effects of Xiaoer Kechuan GranuleObjective:To observe the expectorant effects of Xiaoer Kechuan Granule. Methods: Expectorant effects were evaluated by measuring the excretion of phenol red in mice trachea, after administration of9g/kg,4.5g/kg,2.25g/kg for successive there days.. Results: compared with control group, Xiaoer Kechuan Granule increased the excretion of phenol red from mice trachea (P<0.05or P<0.01). Conclusion:Xiaoer Kechuan Granule showed certain expectorant effects.3. Study on anti-asthmatic effects of Xiaoer Kechuan Granule through guinea pig test induced by Ach and HA.Objective:To observe anti-asthmatic effects of Xiaoer Kechuan Granule. Methods: Anti-asthmatic actions in guinea pigs were observed by histamine and acetyleholine ultrasonic atomization test. Results:compared with control group, Xiaoer Kechuan Granule at the dosage of6g/kg,3g/kg could obviously prolong the asthmatic latent period through guinea pig test induced by Ach and HA (P<0.05). Conclusion:Xiaoer Kechuan Granule showed certain anti-asthmatic effects.4. Study on effects of Xiaoer Kechuan Granule on the inhibition of increased airway responsiveness through mice test induced by Ach and HA.Objective:To observe the effects of Xiaoer Kechuan Granule on the bronchia smooth muscle. Methods:The test of air-overflow of lungs in rats was used to observe the anti-asthmatic effect. Results:compared with control group, Xiaoer Kechuan Granule inhibited the increase of pressure needed by pneumothorax rats (P<0.05or P<0.01). Conclusion:Xiaoer Kechuan Granule could significantly enlarge the bronchia smooth muscle and reduce the rat airway resistance.5. Study on effects of Xiaoer Kechuan Granule on rat asthma model made by ovalbuminObjective:To observe the effects of Xiaoer Kechuan Granule on rat asthma model. Methods:The rat asthmatic model was established by sensitizing rats with ovalbumin, the changes of the expression levels of IgE、IL-4in serum and the Bronchial alveolar lavage fluid were observed. Results:compared with model group, the granule at the dosage of6g/kg,3g/kg,1.5g/kg could obviously inhibit the increase of EOS (P<0.05or P<0.01), the granule at the dosage of6g/kg and1.5g/kg could remarkably inhibit the expression of serum IgE (P<0.05). Conclusion:Xiaoer Kechuan Granule showed remarkable anti-asthmatic effects. 6. Study on effects of Xiaoer Kechuan Granule on the expression of TNF-α、IL-10in lung injury in rats induced by endotoxin.Objective:To observe anti-inflammatory and immunomodulatory effects of Xiaoer Kechuan Granule. Methods:The animal model of lung injury was established by endotoxin in rats, and the inhibition of respiratory inflammation was measured. Results:After Xiaoer Kechuan Granule was administrated for three days,the granule at the dosage of9g/kg and4.5g/kg could inhibit the expression of TNF-αin lung, compared with model group,(P<0.01). The9g/kg dose group can inhibit the reduction of IL-10in lung (P<0.05). Conclusion:Xiaoer Kechuan Granule could certainly inhibit the respiratory inflammation induced by endotoxin.7. Study on effects of Xiaoer Kechuan Granule on the swelling of hind paw in rats induced by carrageenan and the granuloma in rats induced by cotton pellets.Objective:To observe anti-inflammatory effects of Xiaoer Kechuan Granule. Methods: Anti-inflammatory effects were observed by the swelling of hind paw in rats induced by carrageenan and granuloma in rats induced by cotton pellets. Results:compared with control group, All dose groups could inhibit the paw edema induced by carrageenan (P<0.05or P<0.01), the granule at the dosage of6g/kg and3g/kg could certainly decrease the granuloma induced by cotton pellet in SD rats. Conclusion:Xiaoer Kechuan Granule showed certain anti-inflammatory effects.
