Purpose:To observe the changes of serum IgE,PIP2,IP3,CaN levels and degranulation of dopal mast cells in acupuncture at Quchi and Xuehai points in rats with type I allergy ?urticaria?model,and to explore the therapeutic mechanism of CU prevention and treatment.Provide more theoretical basis and ideas for clinical treatment of CU.Materials and methods:50 SPF SD male rats were randomly fed into a group of 5groups according to the random number table method,which were normal group,model group,acupuncture group?Quchi,Xuehai?,loratadine group and Anti-ovin serum preparation group,10 in each group.Take SD rats in the anti-oval protein serum extraction group,and inject 1%of Bacillus vaccine diluent into the abdominal muscles of the hind legs,0.5ml on each side,12 days later?IgE serum peak?The arteries were bled,the serum was separated and mixed,and stored at-20°C until use.Blank group:feeding at room temperature,regular eating,distilled water,17 days later,and measuring indicators.Model group:room temperature feeding,routine feeding,distilled water,anti-oval protein serum was injected on the 15th day,and secondary antigen challenge was performed after 48 hours.After 1 hour,the material was taken and the index was detected.Acupuncture group:feeding at room temperature,regular feeding,distilled water,acupuncture at Rat“Quchi”?with a sulcus in the lateral aspect of rats'back skin tissue microscopyMicroscopically,basal cell destruction,unclear interface,severe keratinization of the epidermal layer,thickening of the granular layer,severe edema between cells,and various inflammatory cell infiltration in the dermis;compared with the model group,acupuncture group and chlorhexidine The structure of the subcutaneous tissue of the rats in the group had no obvious damage and the structure was clear.2.Changes of mast cells in the subcutaneous connective tissue of the rat:Compared with the normal group,mast cells in the subcutaneous connective tissue of the model group showed massive degranulation of mast cells;compared with the model group,the acupuncture group and the chlorhexidine A small part of mast cells were degranulated in mast cells in the connective tissue of the back skin of rats.3.Comparison of mast cell degranulation rate and inhibition rate in subcutaneous connective tissue of rats:Compared with normal group,there was significant difference in model group?P<0.05?;compared with model group,acupuncture group and loratadine group There were significant differences?P<0.05?.There was no significant difference between the acupuncture group and the loratadine group?P>0.05?.4.Compared with the normal group,the serum IgE levels in the model group were significantly increased?P<0.05?.Compared with the model group,the serum IgE levels in the acupuncture group were significantly lower,and there were significant differences.?P<0.05?;there was no significant difference in serum IgE levels between the acupuncture group and the loratadine group?P>0.05?.5.Compared with the normal group,the serum levels of PIP2,IP3 and CaN in the model group were significantly increased?P<0.05?.Compared with the model group,the serum levels of PIP2,IP3 and CaN in the acupuncture group were compared.There was a significant difference?P<0.05?.There was no significant difference in serum PIP2,IP3 and CaN between the acupuncture group and the loratadine group?P>0.05?.Conclusion:1.Acupuncture at Xuehai and Quchi points can inhibit the inflammatory infiltration of CU lesions and restore the structure of tissues.2.Acupuncture at Xuehai and Quchi points has the effect of inhibiting degranulation of mast cells in tissues of CU inflammation.3.Acupuncture at Xuehai and Quchi points can effectively reduce the content of serum IgE,PIP2,IP3 and CaN,and play a role in the treatment of CU.4.Acupuncture at Xuehai and Quchi may inhibit the degranulation of mast cells by regulating PIP2/IP3/Ca2+signaling pathway,and play a preventive role in CU.
